摘要
目的观察施灸赫依三穴对PCPA模型大鼠行为学及脑干组织中5-HT含量的影响。方法将60只正常雄性SD大鼠按体重随机分为6组,分别为正常组、模型组、安定组、顶会穴组、赫依穴组和黑白际穴组,每组10只。除正常组外其余5组腹腔注射PCPA混悬液(1 mL/100 g),连续注射2 d制造大鼠失眠模型。成功造模后进行干预:正常组常规喂养;模型组每天抓取固定1次,常规喂养;安定组灌胃安定(地西泮)注射液(0.92mg/kg),1次/d,连续灌胃7 d;顶会穴组施灸顶会穴、赫依穴组施灸赫依穴、黑白际穴组施灸黑白际穴,每日施灸1次,每次5 min,连续施灸7 d。通过转棒平衡实验观察对比各组大鼠棒上停留时间,并采用高效液相色谱法(LC-MS/MS)检测大鼠脑干5-HT的含量。结果 1)转棒平衡实验:模型组大鼠在转棒上维持时间缩短,有显著性差异(P <0.05)。与模型组比较,安定组、赫依穴组、顶会穴组、黑白际穴组大鼠在转棒上维持的时间大于模型组(P <0.05)。2)大鼠脑干中5-HT的含量:模型组大鼠脑干中5-HT的含量显著降低(P <0.05)。与模型组比较,安定组、赫依穴组、顶会穴组大鼠脑干中5-HT含量升高,有显著性差异(P <0.05),黑白际穴组脑干5-HT含量无显著性差异。结论施灸赫依穴和顶会穴可通过提高脑干组织5-HT的含量,从而使大脑兴奋性降低,起到改善睡眠作用。施灸黑白际穴对大鼠脑干中5-HT含量无影响,其原因可能与不同穴位所分布的5-HT含量与受体有关。
Objective Observing the effects in PCPA rats behavior and the content of 5-HT in brain stem cells through moxibustion.Methods The 60 normal male SD rats were randomly divided into six groups,as the normal group,the model group,the Diazepam group,the Dinghui group,the Heyi point group and the Black and White point group,each group contents 10 rats.Except the normal group,the other five groups were injectedwith PCPA mixed suspension (1mL/100g)and continuously inject 2days to make the insomnia model rats.Interposing after modeling successfully:the normal group feeds regularly.The model group grabs fixation once a day and is fed regularly.Diazepam group was given Diazepam injection (0.92mg/kg)once a day for 7 days.The Dinghui group has acupuncture on Dinghui point,the Hela group has acupuncture on Hela point,the black and white group has acupuncture on black and white point once a day,5 min each time,7 days in a row.The retention time on the bar was observed and the content of 5-HT in the brain stem of rats was measured by high performance liquid chromatography (LC-MS/MS).Results 1)The bar balance experiment:In the model group,the maintenance time on the rotating rod was shortened,and there was significant difference between the two groups.(P <0.05),which is less than the other groups(P <0.05).2)The content of 5-HT in the brain stem of rats:The contents of 5-HT were significantly reduced in the model group (P <0.05),compared with the model group,the content of 5-HT in the brain stem of the Diazepam group,the Hela group,and the Dinghui of the group was elevated (P <0.05),but there was no significant difference from black and white point group.Conclusion Giving moxibustion on Hela point and Dinghui points can improve the content of 5-HT in brain stem tissue,thus decrease the excitability of brain and improve sleep.Giving moxibustion on black and white acupuncture point is not affected the contents of 5-HT.The reasons may be related to the 5-HT content of the different acupoints.
