四种培养液对萱藻种质复壮的影响 被引量:2

Effects of Four Culture Media on Rejuvenation of Germplasm of Scytosiphon lomentaria

摘要 本实验以2012年采自烟台海域的萱藻种质为材料,选取f/2、F1、MAV#3和ES 4种培养液,每种培养液分别设置4种浓度梯度对萱藻种质进行复壮,并与2015年(当年)的种质对比,研究不同种类和浓度的培养液对萱藻种质复壮的影响。本底实验使用1倍浓度的F1培养液,数据显示2012年萱藻种质存在退化现象,在培养第20d,复壮前的2012年和2015年萱藻种质增重倍比分别为29.91%和56.61%,培养第30d的孢子囊比例分别为10.48%和12.99%。培养液复壮实验的第20d和第30d分别计算增重倍比和孢子囊比例,结果显示:(1)1/4倍浓度的ES培养液最适于对萱藻种质进行复壮,复壮后的萱藻种质增重倍比提高至75.04%,同时孢子囊比例提高至14.43%;(2)1倍浓度的f/2培养液只适用于萱藻种质的丝状体生长,但不适于其孢子囊形成,复壮后的增重倍比为46.88%,孢子囊比例的为8.65%;(3)2倍浓度的F1培养液较适于对萱藻种质进行复壮,复壮后的增重倍比提高至30.58%,孢子囊比例提高至12.69%;(4)MAV#3不适用于萱藻种质的复壮,2倍浓度的MAV#3培养液复壮后的增重倍比和孢子囊比例分别降低为23.19%和7.84%。因此,1/4倍浓度ES培养液对萱藻种质的复壮效果最佳。 In this research,the germplasm of Scytosiphon lomentaria,collected from the coast of Yantai in 2012 and preserved in our laboratory,were used as the experimental materials.The increasing ratio and the sporangium proportion were used as the indicators of the growth and development of the germplasm of S.lomentaria,which were compared with that of 2015’s(the current year’s)germplasm.The rejuvenation experiments selected different concentrations(1/4,1/2,1,2 times)of 4 culture media(f/2,F1,MAV#3,ES)to rejuvenate the germplasm of S.lomentariaof 2012.In the background test,the germplasm of S.lomentariaof 2012 and 2015 were cultured by 1 times concentration of F1.The results showed that the increasing ratio of the germplasm of S.lomentariain 2012 and in 2015,respectively,were 29.91% and 56.61% on the 20 th day;the sporangium proportions were 10.48% and12.99% on the 30 th day.The results of the rejuvenation experiments showed as follows:(1)1/4 times concentration of ES was suitable to rejuvenate the germplasm of S.lomentaria,the increasing ratio was increased to 75.04% on the 20 th day,and the sporangium proportion was increased to 14.43% on the30 th day.(2)1 times concentration of f/2 culture medium could be used for the growth rate of filaments,but could not be suitable for sporangium formation;the increasing ratio was 46.88% on the 20 th day,while the sporangium proportion was 8.65% on the 30 th day.(3)2 times concentration of F1 was helpful to rejuvenate the germplasm of S.lomentaria,the increasing ratio was increased to 30.58% on the 20 th day,and the sporangium proportion was increased to 12.69% on the 30 th day.(4)MAV#3 was not adapted to rejuvenating the germplasm of S.lomentaria,the increasing ratio was reduced to23.19% on the 20 th day;meanwhile the sporangium proportion was reduced to 7.84% on the 30 th day.In conclusion,1/4 times concentration of ES was the optimal culture medium to rejuvenate the germplasm of S.lomentaria.
