摘要 目的:检测生发肽的致突变作用,探讨生发肽的遗传毒性。方法:通过Ames实验,中国仓鼠肺成纤维细胞染色体畸变实验以及小鼠骨髓嗜多染红细胞微核实验,评价生发肽的遗传毒性。结果:在Ames实验中,S9代谢和非代谢活化条件下,生发肽浓度为每皿≤5 000μg,对所测5种菌株回复突变菌落数均未出现剂量依赖性增加;在染色体畸变实验中,S9代谢和非代谢活化条件下,药物浓度为165,330和660μg·mL^(-1)时,细胞染色体畸变率与溶剂对照组相比差异均无统计学意义(P> 0. 05),且无剂量—反应关系;在小鼠骨髓细胞微核实验中,在250,500和1 000 mg·kg^(-1)3个剂量组中均未见骨髓中含微核的嗜多染红细胞数增加。结论:在实验剂量范围内,生发肽遗传毒性结果为阴性。 Objective:To investigate the mutagenicity of germinal peptide and explore its genetic toxicity. Methods:The Ames test,chromosomal abnormality test of Chinese hamster lung cells,and micronucleus test of mouse bone marrow polychromatic erythrocytes were carried out to evaluate the genetic toxicity of germinal peptide. Results:In the Ames test,under the conditions of S9 metabolism and non-metabolic activation,the numbers of five strains of revertant colonies did not show dose-dependent increase when the germinal peptide concentration was under 5000μg·plate^-1 In the chromosome aberration test,in the S9 metabolic and non-metabolic activation conditions,when the concentrations of germinal peptide were 165,330 and 660μg·mL^-1,there was no significant difference in chromosome aberration rate compared with thoes in the control group (P>0.05),and there was no dose-response relationship.In the micronucleus test of mouse bone marrow cells,there was no increase in the number of micronucleated polychromatic erythrocytes in the bone marrow at any of the three doses of 250,500 and 1000mg.kg^-1.Conclusion:In the range of experimental dose,the result of genotoxicity of germinal peptide was negative.
