EHD2低表达对肝癌预后的影响

Effect of Low Expression of EHD2 on Prognosis of Hepatocellular Carcinoma

摘要 目的:研究EHD2(EH domain-containing protein-2)在肝细胞肝癌(hepatocellular carcinoma,HCC)组织中的表达及临床意义。方法:应用免疫组织化学方法检测EHD2在肝细胞肝癌组织及其癌旁组织中的表达情况,并分析其表达和临床病理特征、患者预后之间的关系。结果:EHD2蛋白在肝细胞肝癌组织中的表达显著高于对应癌旁组织,EHD2的表达和性别、组织学分级、肿瘤转移、侵袭相关(P<0.05)。Kaplan-Meier生存率曲线表明EHD2高表达的HCC患者总生存率和无瘤生存率显著延长。多因素分析提示EHD2是肝细胞肝癌患者一个独立的预后因子。结论:EHD2在HCC组织中高表达,且和肿瘤的组织学分级呈正相关,和肿瘤转移、侵袭呈负相关,提高肝癌患者总生存率和无瘤生存率,是HCC患者一个独立的预后因子,有望成为HCC治疗新的靶点。 Objective To investigate the expression and clinical significance of EHD2( EH-domain containing protein 2) in hepatocellular carcinoma( HCC). Methods EHD2 expression levels in the HCC specimens and adjacent noncancerous tissues were detected by immunohistochemistry. The correlation of EHD2 expression with the clinicopathological features and prognosis of patients was analyzed. Results The expression of EHD2 was significantly higher in HCC tissues than that in adjacent normal tissues. The EHD2 expression was correlated with sex,histological grade,tumor metastasis and microvascular invasion( P<0.05). The Kaplan-Meier survival curve showed that the overall survival and tumor-free survival rates of HCC patients with high EHD2 expression were significantly prolonged. Multivariate analysis suggested that EHD2 is an independent prognostic factor in patients with HCC. Conclusion EHD2 was highly expressed in HCC tissues and positively correlated with tumor histological grade,and negatively correlated with tumor metastasis and invasion. It is an independent prognostic factor for HCC patients and is expected to become a new target for HCC treatment.
