摘要
目的运用SCoT分子标记技术对广东化州化橘红进行种内品系间的亲缘关系分析研究。方法通过提取化橘红DNA,正交设计寻找化橘红SCoT分子标记最佳PCR反应体系,筛选获得化橘红PCR扩增的SCoT引物,应用NTSYS2.10e软件并采用UPGMA法进行聚类分析。结果发现SCoT引物碱基序列共扩增出81条带,其中多态性条带有44条,种内品系间的相似系数在0.75~1之间,不同品系化橘红分别聚为一类。结论表明SCoT分子标记可以有效运用于化橘红的种内亲缘关系分析和种内品系鉴定研究。
Objective To investigate the genetic relationship of Exocarpium Citri Grandis in Huazhou by using SCoT molecular marker technology. Methods The DNA of Exocarpium Citri Grandis is extracted and the best condition for PCR reaction is sought out by orthogonal design method. SCoT primers are screened out for PCR amplification and the UPGMA cluster analysis is carried out by NTSYS2.10 e software. Results81 bands are amplificated out by the SCoT primer base sequence, including 44 polymorphic bands. The intraspecific similarity coefficient is between 0.75~1.00. Exocarpium Citri Grandis of different varienties are respectively divided into category. Conclusion All the results suggest that SCoT could be a molecular marker for the relativety relationtion and the genetic diversity of Exocarpium Citri Grandis species.
