寨卡病毒亚洲型与非洲型基因同源性及重组分析 被引量:1

Homology and recombination of Asian and African genetypes of Zika virus genome

摘要 目的分析寨卡病毒(Zika virus,ZIKV)亚洲型与非洲型基因组核苷酸序列同源性及结构蛋白C、前膜蛋白(pre-membrane,pr M)核苷酸和氨基酸序列差异,并探讨了ZIKV亚洲型和非洲型位点突变可能产生的后果。方法在GenBank中登录的ZIKV亚洲型和非洲型序列中,分别选择5个不同的全长基因组序列,并获得其相应的氨基酸序列,采用BIOEDIT软件进行比对,分析相似度,统计不同型别间结构蛋白C、prM的核苷酸突变和氨基酸替代位点,并采用SimPlot软件分析两个型别间的同源重组。结果 ZIKV亚洲型与非洲型的核苷酸相似性在88. 00%~89. 00%之间;不同型别的结构蛋白C和prM碱基的突变比较显示,C基因共有19个碱基突变,prM基因共有48个碱基突变;氨基酸翻译比对显示,这67个碱基突变均为有效突变。两个型别结构蛋白C和prM编码的289个氨基酸中,占组成比例最多的均为亮氨酸(L),同源重组分析亚洲型与非洲型一致性较高,无重组信号。结论通过对ZIKV亚洲型与非洲型的结构蛋白C、pr M核苷酸序列、氨基酸的比较分析及同源重组分析,为研究ZIKV不同基因型间的生物学和致病性差异提供参考。 Objective To analyze the homology of nucleotide sequences as well as the differences in nucleotide and amino acid sequences of structural protein C and pre-membrane (prM)protein of Asian and African genotypes of Zika virus (ZIKV)genome,and investigate the possible result of site-directed mutation of the two genotypes.Methods Five full-length genome sequences of Asian and African genotypes of ZIKV were collected from GenBank,of which the amino acid sequences were compared by using BIOEDIT software to analyze the homology.The nucleotide mutation and amino acid substitution sites of structural protein C and prM protein of the two genotypes were recorded to analyze the homologous recombination by using SimPlot software.Results The homology of nucleotides of Asian and African genotypes of ZIKV was 88.00%-89.00%.A total of 19 base mutations were observed in C gene while 48 in prM gene, all of which were nonsynonymous mutation.The structural protein C and prM of both genotypes encoded 289 amino acids, of which leucine (L)accounted for the largest proportion of components.Asian genotype was highly homologous to African genotype,and no recombinant signal was found.Conclusion The comparative analysis of nucleotides and amino acids of Asian and African genetypes of ZIKV virus genome as well as homologous recombination provided a reference for study on the difference in the biology and pathogenicity of ZIKV of various genotypes.
