产后缺乳肝郁脾虚病证结合大鼠模型的建立与评价 被引量:7

Establishment and Review of Rat Model of Postpartum Hypogalactia with Syndrome of Liver Depression with Spleen Insufficiency

摘要 目的:探索产后缺乳肝郁脾虚病证结合大鼠动物模型的构建方法。方法:使用甲磺酸溴隐亭激活多巴胺受体抑制催乳素分泌,建立大鼠产后缺乳模型;采用慢性束缚致大鼠肝郁、肝疏泄不及、肝郁横逆犯脾致肝郁脾虚,建立产后缺乳中医证候模型;观察大鼠每小时泌乳量、仔鼠窝净增重、大鼠体质量、进食进水量、大鼠宏观表征、旷场实验、血清CRH、ACTH、COR、GAS含量变化情况。结果:模型组大鼠的每小时泌乳量、大鼠体质量、进食进水量均明显低于对照组(P<0.05),模型组仔鼠窝净增重低于对照组(P<0.05)。大鼠宏观表征造模前后差异明显。旷场实验中模型组大鼠水平运动、垂直运动次数均少于对照组(P<0.05)。模型组大鼠血清CRH、ACTH、COR含量均明显高于对照组(P<0.05),而GAS含量明显低于对照组(P<0.05)。结论:模拟西医病因病理、传统中医病机建立产后缺乳动物模型,并通过动物宏观表征、临床与实验室指标等方法综合评价可成功构建产后缺乳大鼠病证结合动物模型。 Objective:To establish a rat model of postpartum hypogalactia with syndrome of liver depression with spleen insufficiency.Methods:The postpartum hypogalactia rat model was established to simulate the etiology and pathology of disease.Bromocriptine mesylate was utilized to activate dop amine receptor to inhibit prolactin secretion.The animal model of postpartum hypogalactia of syndrome of liver depression with spleen insufficiency was established by chronic restraint method during the process.Chronic restraint method caused liver stagnation and spleen depression.The rat model was reviewed in following aspects:Lactation volume per hour of rats;Net weight gain in the litter;The weight,food intake and water intake of rats;Macroscopic representation of rats;Open field experiment;The hypothalamic pituitary adrenal axis (The hypothalamic-pituitary-adrenal axis, HPA axis)related indicators:plasma corticotropin releasing hormone (CRH),plasma adrenocorticotropic hormone (ACTH),plasma cortisol (COR);Plasma GAS.Results:The Lactation volume per hour,the weight of rats,the food and water intake of the model group were significantly reduced compared with that of the control group (P<0.05).The net weight gain in the litter of the model group was slower than that of the control group (P< 0.05).There were obviously differences of the macroscopic representation in the comparison.In the open-field experiment,both horizontal and vertical movements of rats in the model group were reduced compared with those in the control group (P<0.05).Compared with the control group,plasma COR,CRH and ACTH in the model group increased significantly (P<0.05)and GAS level decreased (P<0.05).Conclusions:By simulating the etiology and pathology of Western medicine and the pathogenesis of traditional Chinese medicine,and through the comprehensive evaluation of animal macro-characterization,clinical and laboratory indicators,we can successfully construct the animal model of postpartum hypogalactia combined with disease and syndrome.
