臭氧膀胱灌注对膀胱过度活动症大鼠膀胱黏膜P2X3的影响 被引量:3

Effect of Ozone-water Intravesical Instillation on Bladder Mucosal P2X3 in Rats with Overactive Bladder

摘要 目的:评价臭氧水膀胱腔内灌注疗法对膀胱过度活动症(OAB)大鼠P2X3受体的影响。方法:通过建立膀胱过度活动症大鼠动物模型,尿流动力学检测证实膀胱逼尿肌不稳定,然后将大鼠共分为3组:正常组、膀胱过度活动症组(OAB组)和臭氧治疗组,然后分别采用Masson染色了解膀胱结构情况,通过Western Blot测定三组大鼠膀胱黏膜P2X3蛋白量变化,获得的数据采用t检验进行统计学分析。结果:共获取26只膀胱过度活动症大鼠模型,采用臭氧水膀胱腔内灌注处理13只(臭氧治疗组),13只作为OAB组,尿流动力学实验正常的大鼠作为正常组(14只)。三组大鼠膀胱黏膜P2X3表达有差异(P<0.05),使用臭氧水膀胱灌注后,P2X3受体表达量下降,同灌注前有差异(P<0.05)。结论:臭氧水灌注治疗能改善膀胱过度活动症大鼠的膀胱功能,机制可能为臭氧水膀胱腔内灌注疗法能降低膀胱过度活动症大鼠P2X3受体表达。 Objective To evaluate the effect of ozone-water intravesical instillation on P2X3 receptor expression in rats with rat model of overactive bladder(OAB).Methods Rat model of overactive bladder was induced by bladder outlet obstruction (BOO)and confirmed by the proof of bladder detrusor instability through urodynamic testing.Rats were divived into control group,OAB group and ozone treatment group.Masson staining was done in rat bladder tissue to evaluate the structure changes.Western blot was performed to examine the expression of P2X3 in mucosa of bladder in all groups.The.data were analyzed by t test.Results A total of 26 rat models of overactive bladder were obtained.Thirteen rats were treated with intravesical instillation of ozone-water,13 were used as OAB group,and rats with normal urodynamics were used as normal control (n=14).The expression of P2X3 in rat bladder mucosa of the three groups was different (P<0.05).After intravesical instillation with ozone water,the expression of P2X3 receptor decreased,which was different from that before perfusion (P<0.05).Conclusion Ozone water intravesical instillation can improve bladder function in rats with overactive bladder. The mechanism may be that ozone water intravesical instillation therapy can reduce the expression of P2X3 receptor in rats with overactive bladder.
