从变形杆菌蔓延生长中分离气单胞菌的方法探讨

摘要 目的了解不同培养基对变形杆菌、气单胞菌的生长情况的影响,为2种细菌在混合感染时分离纯化气单胞菌提供方法依据。方法选取实验室常用平板培养基分别划线接种变形杆菌和气单胞菌进行分离培养,观察2种细菌在不同平板上菌落生长形态和生化反应结果。结果实验室常用普通培养基对变形杆菌蔓延生长无抑制作用;接种SS、TCBS、EMB、沙门菌显色培养基、弧菌显色培养基能够抑制变形杆菌蔓延生长,但对气单胞菌生长有抑制作用; XLD平板能够有效抑制变形杆菌蔓延生长,变形杆菌在XLD平板上呈现中等大小、黄色、中心有黑色或无黑色菌落,而XLD平板对气单胞菌无抑制,培养基上出现中等偏大、无色半透明的扁平状菌落。结论变形杆菌和气单胞菌混合生长时,接种XLD平板得到2种不同生长形态的菌落,从而可以有效分离2种菌。 Objective To explore the effect of different medium on the growth of Aeromonas and Proteus,so as to provide basis for the separation and purification of Aeromonas in mixed infection.Methods The two kinds of bacteria were inoculated and cultured in different kinds of common plating medium,and then the colony morphologies and biochemical reaction were observed.Results Ordinary medium had no inhibitory effect on the spread of Proteus;inoculations of SS,TCBS,EMB, Salmonella chromogenic medium,Vibrio chromogenic medium can inhibit the spread of Proteus,but had inhibitory effect on the growth of Aeromonas;XLD plates can effectively inhibit the spread of Proteus,and medium-sized,yellow,black or black colonies were found on XLD plates,while XLD plates had no inhibition on Aeromonus,and medium to large and colorless trans-lucent fiat colony was observed.Condusion Inoculating XLD on plates can separate Aeromonus from Proteus when mixed spreading occurs.
