摘要
建立了一种柱前衍生高效液相色谱法测定市售护发产品中赖氨酸含量的方法。样品经0.1 mol/L盐酸溶液超声提取,经设定好的进样程序在线衍生后,采用安捷伦ZORBAX Eclipse XDB-C18柱(250 mm×4.6 mm×5μm)进行分离。选用V(乙腈)︰V(甲醇)︰V(水)=45︰45︰10的有机相和40 mmol/L磷酸二氢钠缓冲溶液(pH=7.8)作为流动相进行梯度洗脱,检测波长为338 nm,柱温是25℃。结果表明:赖氨酸标准溶液在10~500 mg/L的质量浓度范围内保持良好的线性关系,相关系数R大于0.99,检出限为5.0 mg/kg,稳定性和重复性良好,加标回收率在96.42%到111.24%之间,相对标准偏差3.91%~5.23%之间。该方法操作简便,可用于护发产品中赖氨酸含量的测定。
The method for determining the content of lysine in hair care products by pre-column derivatization of HPLC was established. The samples were extracted with 0.1 mol/L HCl by ultrasonic technique. After derivatization with on-line pre-column procedure, they were separated by Aglient ZORBAX Eclipse XDB-C18 column(250 mm×4.6 mm×5 μm). The mobile phases were organic phase of ACN-MeOH-Water(volume ratio of 45/45/10) and 40 mmol/L NaH2 PO4(pH=7.8). The wavelength of UV-detection was 338 nm and the temperature of column was 25 ℃. The results showed that the lysine standard solution had good liner relationship in range of 10 to 500 mg/L with correlation coefficient more than 0.99. The test had good stability and reproducibility with detection limit of 5.0 mg/kg, the recovery and RSD were in the range of 96.42% to 111.24% and 3.91% to 5.23%, respectively. The method is convenient for determining the content of lysine in hair care products.
