摘要
目的探讨miR-497及其靶基因ANLN编码的Anillin蛋白在鼻咽癌发展中的作用。方法采用qPCR和免疫组化方法分析鼻咽癌组织及正常鼻咽部组织中miR-497和Anillin蛋白的表达情况;多因素Cox回归分析其与预后的关系;将ANLN-m RNA转染至鼻咽癌细胞,通过Tra nswell侵袭实验观察其对鼻咽癌细胞侵袭的影响。结果 miR-497在鼻咽癌组织中相对于正常鼻咽部组织低表达;ANLN及Anillin蛋白在鼻咽癌组织中高表达;多因素Cox回归分析显示miR-497表达可以作为患者预后的独立影响因素;ANLN-mRNA转染HONE1及HNE1细胞穿过小室的数值分别为162.89±12.32、139.76±7.85,对照组为68.37±9.83、82.32±5.96,差异均有统计学意义(P均<0.05)。结论 miR-497在鼻咽癌组织中表达下降,具有抑癌基因作用;miR-497调控靶基因ANLN编码Anillin蛋白影响鼻咽癌增殖及侵袭。
OBJECTIVE To investigate the roles of miR-497 and its target gene ANLN encoded Anillin protein in the development of nasopharyngeal carcinoma(NPC).METHODS The expression of miR-497 and Anillin in NPC and normal nasopharyngeal tissue were detected with qPCR and immunohistochemistry methods. Multivariate Cox regression analysis was used to analyze the relationship between expression of miR-497 and Anillin and prognosis.ANLN-mRNA was transfected into NPC cells and its effect on invasion of NPC cells was observed by Transwell invasion test. RESULTS MiR-497 expression was lower in NPC tissues compared with normal nasopharyngeal tissue. ANLN and its protein Anillin were over expressed in NPC tissues. Multivariate cox regression analysis showed that miR-497 was an independent prognostic factor for NPC. The number of ANLN-mRNA transfected HONE1 and HNE1 cells passing through the compartments was162.89±12.32 and 139.76±7.85 respectively, while that of the control group was 68.37±9.83 and 82.32±5.96 respectively.The difference was statistically significant(P <0.05).CONCLUSION The expression of miR-497 is decreased in NPC, it has the function of tumor suppressor gene. miR-497 may modulate NPC cell growth and invasion through Anillin protein encoded by gene ANLN.
