白藜芦醇对甲状腺癌SW579细胞株的抗肿瘤作用及机制探讨 被引量:4

摘要 目的观察白藜芦醇对甲状腺癌SW579细胞株的抗肿瘤作用并探讨可能作用机制。方法体外常规培养甲状腺癌SW579细胞,将细胞分为对照组和白藜芦醇组(终浓度为20、40、80μmol/L),MTT法检测甲状腺癌SW579细胞活性,BrdU荧光染色检测甲状腺癌SW579细胞增殖,DAPI染色法检测细胞凋亡,caspase-3和caspase-9活性检测试剂盒分析白藜芦醇对甲状腺癌SW579细胞亡的作用机制。结果白藜芦醇(20、40、80μmol/L)组甲状腺癌SW579细胞活性、BrdU表达、DA PI染色显著低于对照组,但细胞内caspase-3和caspase-9活性上升(P<0.01)。Ac-DEVD-CHO和ZLEHD-FMK可显著提高白藜芦醇组(80μmol/L)中甲状腺癌SW579细胞的活性(P<0.05)。结论白藜芦醇对抗甲状腺癌细胞具有抑制作用,通过激活caspase-3和caspase-9内源性凋亡信号通路可能是其作用机制之一。 OBJECTIVE To observe the anti-tumor effect of resveratrol on thyroid carcinoma SW579 cell line, and investigate its mechanism.METHODS Thyroid cancer SW579 cells were cultured according to the conventional method.The experiment was divided into control group and resveratrol group (20,40,80μmol/L).MTT assay was used to detect the cell viability of thyroid carcinoma SW579 cell line,BrdU fluorescent staining to detecte the proliferation of thyroid cancer cells,DAPI staining to detect apoptosis,and activity detection kit of caspase-3 and caspase-9 to analyze the mechanism of effect of resveratrol on the cell death of thyroid carcinoma SW579 cell line.RESULTS Compared with control group,the activity of SW579 cells of thyroid carcinoma,the expression of BrdU(optical density,surface density)and DAPI staining(optical density,surface density) of resveratrol groups(20,40,80μmol/L)were significantly decreased while activity of caspase-3 and caspase-9 in thyroid carcinoma SW579 cell line were remarkably increased(P<0.01).Ac-DEVD-CHO and Z-LEHD-FMK significantly increased the cell viability of thyroid carcinoma SW579 cell line treated with resveratrol (80μmol/L). CONCLUSION Resveratrol has the anti-tumor effect for thyroid carcinoma SW579 cell line,and activation of the endogenous apoptotic signaling pathways of caspase-3 and caspase-9 may be one of its mechanisms.
