UPLC-UV法同时测定葛酮通络胶囊中的6种活性成分

Simultaneous Determination of Six Kinds of Active Constituents in Getong Tongluo Capsules by UPLC-UV

【目的】应用超高液相色谱—紫外(UPLC-UV)法同时测定葛酮通络胶囊中6种活性成分。【方法】采用WatersACQUITY UPLC系统,BEH Shielded RP-C18色谱柱(2.1 mm×100 mm,1.7μm),柱温35℃,流动相采用乙腈和0.1%甲酸水,流速为0.2 mL·min-1。紫外检测波长为250 nm。【结果】所选6种活性成分可以同时实现分离并定量,所有6种成分的标准曲线的相关系数都大于0.998。6种成分的平均加样回收率及相对标准偏差(s R)分别为:3’-羟基葛根素,100.29,1.21%;葛根素,100.11,1.40%;大豆苷,99.73,1.15%;染料木苷,99.93,1.24%;大豆苷元,100.99,1.31%;染料木素,99.68,1.04%。【结论】本方法准确、快速、重复性好,可以作为全面控制葛酮通络胶囊质量的新方法。

Objective To establish a method for the simultaneous determination of 6 kinds of effective compounds in Getong Tongluo Capsules by ultra-performance liquid chromatography-uhraviolet(UPLC-UV).Methods The chromatography was performed with Waters ACQUITY UPLC system on a BEH shielded RP-C18 column (2.1mm× 100mm,1.7μm)at temperature of 35℃.The mobile phase consisted of acetonitrile and 0.1% formic acid with the flow rate of 0.2mL·min^-1.The UV detection wavelength was 250nm.Results All of 6 kinds of active compounds were simultaneously separated and quantified,and their correlation coefficients in calibration curves were more than 0.998.Their average recovery and relative standard deviation(RSD,SR)of the 6 kinds of active compounds werelistedasfollows:3'-hydroxypuerarin,100.29,1.21%;puerarin,100.11,1.40%;daidzin, 99.73,1.15%;genistin,.99.93,1.24%;daidzein,100.99,1.31%;genistein,99.68,1.04%.Conclusion The validated UPLC-UV is a new method for the quality control of Getong Tongluo capsules,which is accurate, rapid,and reproducible.
