列当多糖对鸡淋巴细胞体外增殖能力的影响

Effect of Polysaccharides in Orobanche caerulescens on Chicken Lymphocyte Proliferative Ability in Vitro

为探讨列当多糖(OSP)对鸡脾脏淋巴细胞和外周血淋巴细胞增殖作用的影响,试验用水提醇沉法提取列当多糖,通过苯酚硫酸法和红外光谱对列当多糖进行检测,采用MTT法测定列当多糖的安全浓度及对鸡淋巴细胞的增殖能力。结果显示:试验所提取的列当多糖均具有多糖的特征峰为多糖类化合物。外周血T淋巴细胞增殖试验表明,OSP30和OSP80的增殖作用强且对T淋巴细胞增殖的浓度范围最大。脾脏B淋巴细胞增殖试验表明,OSP30和OSP80的增殖作用强且对B淋巴细胞增殖的浓度范围最大。结果表明,OSP30及OSP80对脾脏淋巴细胞和外周血T淋巴细胞的增殖作用最强。

To investigate the effect of polysaccharides in Orobanche caerulescens(OSP)on proliferation of lymphocytes andperipheral lymphocytes in chicken spleen. The polysaccharides were extracted by water extraction and ethanol precipitation.The polysaccharides were detected by phenol sulfuric acid and infrared spectroscopy. The safe concentration of thepolysaccharides and proliferative capacity of chicken lymphocytes were determined by MTT method. The results showedthat the characteristic peaks of polysaccharides extracted from the experiment were polysaccharides. The proliferation ofperipheral blood T lymphocytes showed that OSP30 and OSP80had strong proliferation effect and the concentration rangeof proliferation of T lymphocytes was the largest. Splenic B lymphocyte proliferation test showed that OSP30 and OSP80hadstrong proliferation effect and the concentration range of proliferation of B lymphocyte was the largest.The results showed thatOSP30 and OSP80had the strongest effect on the proliferation of splenic lymphocytes and peripheral blood T lymphocytes.
