摘要 本文提出了一种基于聚多巴胺(PDA)涂层锆系金属有机骨架(UiO-66)的新型生物传感器用于三磷酸腺苷(ATP)的检测技术。此技术是由PDA/UiO-66复合纳米材料、羟基荧光素(FAM)分子标记的ATP适配体(P)和脱氧核糖核酸酶I(DNase I)组成的酶辅助信号放大检测系统。PDA/UiO-66复合材料的强荧光猝灭性能使其成为一种高效率、高灵敏度的生物传感器,它的检测限为35 nM。该纳米复合材料的聚多巴胺层增强了此传感器的生物相容性和生物活性,使得其在未来对实际血清样本具有临床诊断的潜力。 In this work,a novel and sensitive biosensor based on polydopamine coated Zr-based metal-organic framework (PDA/UiO-66) is presented for adenosine triphosphate (ATP)detection.This nuclease-assisted signal amplified analysis system consists of PDA/UiO-66 complexes,6-carboxyfluorescein (FAM)-labeled aptamers and DNase I.In the absence of ATP,the aptamers are protected from DNase I due to the aptamers are adsorbed on the surface of PDA/UiO-66 complexes and then the flurescence of FAM is quenched.When ATP molecules exist,aptamers are desorbed from PDA/UiO-66 and cleaved by DNase I. After digestion,the released ATP molecule can hybridize with another aptamer,then the complex structure deviates from the nanosphere and restarts a new recycle.Herein,strong fluorescence quenching ability and uploading more amount of aptamer probes of PDA/UiO-66 nanocomposites make them efficient biosensors, leading to a high sensitivity with detection limit of 35nM.Moreover,the enhanced biocompatibility and bioactivity with PDA layer of the nanocomposites render the proposed strategy to hold potential for clinical diagnosis for detecting ATP in the near future.
