人参皂苷Rg1对MPTP诱导的帕金森病小鼠黑质caveolin-1表达的影响 被引量:2

摘要 目的探讨Cav-1蛋白在帕金森病小鼠黑质表达情况及人参皂苷Rg1对其表达的影响及意义。方法将30只C57BL/6小鼠随机分为正常对照组(C组)、模型组(M组)、人参皂苷Rg1组(R组)。C组和M组小鼠腹腔注射生理盐水,R组小鼠腹腔注射人参皂苷Rg1预防给药,腹腔注射MPTP构建帕金森病模型。采用RT-PCR和Western blotting检测小鼠黑质Cav-1 mRNA和蛋白表达水平。结果M组小鼠黑质Cav-1 mRNA表达水平显著高于C组和R组(P<0.01)。M组小鼠黑质Cav-1蛋白表达水平显著高于C组和R组(P<0.01)。结论 Cav-1蛋白在帕金森病小鼠黑质区呈高表达,人参皂苷Rg1可能通过降低Cav-1蛋白的表达保护和修复多巴胺能神经元。 Objective To explore the influence and significance of Ginsenoside Rg1 on expressions of Cav-1 in substantia nigras of mice with Parkinson 's disease. Methods Thirty C57 BL/6 mice were randomly divided into control group( C group),model group( M group),Ginsenoside Rg1 group( R group). The mice in C group and M group were given physiological saline by intraperitoneal injection while the mice in R group were pretreated with Ginsenoside Rg1 by intraperitoneal injection. MPTP was injected intraperitoneally to mice to build PD model. The expression of Cav-1 protein and mRNA in the substantia nigra of mice was investigated by RTPCR and Western blotting,respectively.Results The level of Cav-1 mRNA expression in the substantia nigras of M group was significantly higher than that of the R group and C group( P < 0.01). The level of Cav-1 protein expression in the substantia nigras of M group was significantly higher than that of the R group and C group( P<0.01). Conclusions MPTP may induce Parkinson ' s disease by promoting the expressions of Cav-1 in the substantia nigras of mice. Ginsenoside Rg1 may improve the symptoms of Parkinson 's disease by reducing the expressions of Cav-1 protein in the substantia nigras of mice.
