摘要
以鸡白痢沙门菌C79-13为宿主菌,从粪便样品中分离到1株裂解性噬菌体SP17,并对获得的噬菌体进行纯化鉴定及生物学特性研究。结果表明:SP17噬菌斑清晰透亮,空斑直径达到5mm;最佳感染复数为0.01;该长尾噬菌体由正多面体头部(直径约60nm)和无收缩性尾巴(长约110nm)组成,对肠炎沙门菌、鸡白痢沙门菌、德尔卑沙门菌及部分鼠伤寒沙门菌具有良好的裂解能力;生长曲线表明噬菌体SP17的潜伏期为10min,爆发期约为80min,爆发量为80PFU/cell;在温度30~60℃、pH值4~12条件下具有良好的稳定性。这一研究提示噬菌体SP17具有应用于沙门菌防控的前景。
A virulent bacteriophage SP17 was isolated from fecal samples by double-layer agar culture method.The plaque of SP17 was round and transparent,and its size was about 5 mm in diameter.SP17 belongs to the family Siphoviridae which had an icosahedral head approximately 60 nm in diameter and a long noncontractile tail with 110 nm in length,and it can specifically infect Salmonella Enteritidis,Salmonella Pullorum,Salmonella Derby,and Salmonella Typhimurium.Its optimal MOI was 0.01.One-step growth curve showed that the latent time and burst time were 10 min and80 min,the burst size was 80 PFU/cell.This bacteriophage SP17 was stable under various pH(4-12)and temperature(30-60℃).These characteristics suggested that SP17 had potential application value for controlling Salmonella.
