摘要
目的观察芦丁对高脂饮食诱导的SAMP8小鼠血脂、糖耐量(GTT)、胰岛素耐量(ITT)及脂肪组织代谢相关蛋白的影响。方法将24只SAMP8小鼠随机分为低脂饲料(LFD)、高脂饲料(HFD)、高脂+芦丁(HR)共3组,每组8只。饲养18w后,进行糖耐量和胰岛素耐量实验,然后称重并处死,取血清测定甘油三酯、总胆固醇和甘油水平,取皮下和附睾脂肪组织用于蛋白检测。结果与LFD组相比,HFD组小鼠附睾脂肪组织重量、GTT和ITT血糖曲线下面积(AUC)显著增加(P<0.05)。HR组小鼠ITTAUC较HFD组显著降低(P<0.05)。在皮下和附睾脂肪组织中,与LFD组相比,HFD组P-Aktser473和P-AMPK蛋白表达量均显著降低(P<0.05);HR组皮下脂肪组织P-AMPK和附睾脂肪组织P-Aktser473较HFD组显著升高(P <0.05);各组间TNFα蛋白表达量无显著差异(P>0.05)。结论芦丁能够改善高脂饲养SAMP8小鼠胰岛素耐量受损状况。且芦丁干预能够升高脂肪组织P-Aktser473和P-AMPK蛋白表达,但是随脂肪组织部位不同而存在差异。
Objective To explore the effect of rutin on blood lipid, glucose tolerance test(GTT), insulin tolerance test(ITT) and adipose tissue metabolism related proteins in SAMP8 mice fed a high fat diet. Methods Totally 24 SAMP8 mice were randomly divided into three groups: the low fat diet group(LFD), the high fat diet group(HFD), and the high fat diet + rutin group(HR), with 8 mice in each group. The glucose tolerance test and insulin tolerance test were conducted in mice after feeding for 18 weeks. Then, the mice were weighted and executed. The serum was collected for detecting the levels of triglycerides, total cholesterol and glycerol, and the subcutaneous adipose tissue and epididymal adipose tissue were collected and stored for further protein analysis. Results The weight of epididymal adipose tissue, as well as the area under the curve(AUC) of blood glucose of GTT and ITT in mice of HFD group were significantly increased compared with those in LFD group(P<0.05). The ITT AUC of mice in HR group was remarkably decreased compared with that in HFD group(P<0.05). Compared with LFD group, the expression of P-Aktser473 and P-AMPK in the subcutaneous adipose tissue and epididymal adipose tissue of mice in HFD group were dramatically reduced(P<0.05);the expression of P-AMPK in the epididymal adipose tissue as well as the expression of P-AMPK in the subcutaneous adipose tissue of mice in HR group were obviously improved compared with those in HFD group(P<0.05);the difference among groups in the expression level of TNFα protein was not statistically significant(P >0.05). Conclusion Rutin improves the impaired insulin tolerance in SAMP8 mice fed a high fat diet. Rutin intervention is capable of increasing the expression levels of P-Aktser473 and P-AMPK. However, their expressions are varied with different adipose tissue depots.
