水果中测定β-胡萝卜素的皂化及直接提取方法比较

COMPARISON OF TWO METHODS FOR DETERMINATION OF Β-CAROTENE IN FRUITS BY SAPONIFICATION AND DIRECT EXTRACTION

摘要 目的研究比较皂化法和直接提取法两种不同前处理方法测定水果中β-胡萝卜素含量。方法以福建省30种本地水果为检测对象,采用优化后的直接提取法和皂化法测定β-胡萝卜素含量,分析比较二者的测定结果及可操作性等。结果皂化法的平均回收率为93.1%~96.2%,相对标准偏差为6.9%~8.4%;直接提取法的平均回收率为73.2%~80.4%,相对标准偏差为2.8%~4.2%。皂化法的平均回收率高于直接提取法。两种方法所测定大部分水果的含量结果有明显的统计学差异(P<0.001),建阳桔柚等4种柑橘类水果含量测定结果相差不大(P>0.05)。结论水果的β-胡萝卜素含量测定中,虽然皂化法实验操作稍微繁杂、耗时,但其测定结果较为准确。 Objective To compare saponification and direct extraction in determining β-carotene content in fruits. Methods Thirty kinds of native fruits in Fujian province were used as test samples. β-carotene content was determined by optimized direct extraction method or saponification method, and the results of analysis were compared. Results The average recoveries by saponification were from 93.1% to 96.2% with RSDs from 6.9% to 8.4%. The average recoveries by direct extraction were from 73.2% to 80.4% with RSDs from 2.8% to 4.2%. The average recovery rate of saponification method was higher than that of the direct extraction method. The concentrations of β-carotene in Jianyang orange grapefruits measured by the two methods were similar(P >0.05), but most other fruits showed significant statistical difference in β-carotene content(P<0.001). Conclusion Although the saponification method is a little complicated and time-consuming, it is more accurate in determining the content of β-carotene in fruits.
