子痫前期产妇胎盘组织中生长阻滞特异性蛋白6、富集AT序列的特异性结合蛋白1的含量及其与氧化应激、炎性反应的相关性 被引量:4

Contents of Gas6 and SATB1 in placental tissue of preeclampsia women and their correlations with oxidative stress and inflammatory reaction

摘要 目的探讨子痫前期产妇胎盘组织中生长阻滞特异性蛋白6 (Gas6)、富集AT序列的特异性结合蛋白1(SATB1)的含量及其与氧化应激、炎性反应的相关性,为临床诊治提供参考依据。方法收集2014年7月-2017年2月在武功县人民医院进行分娩的子痫前期产妇90例作为子痫前期组,同期在武功县人民医院进行分娩的健康产妇50例作为正常对照组。留取两组产妇的胎盘标本组织,采用Western-blot法检测其中Gas6、SATB1 mRNA的表达量;采用酶联免疫吸附法(ELISA)检测胎盘标本组织碾磨液中氧化应激、炎性反应指标的含量。结果子痫前期组产妇胎盘组织标本中Gas6 mRNA表达量高于正常对照组产妇,SATB1 mRNA表达量低于正常对照组产妇,差异有统计学意义(均P<0. 05);子痫前期组产妇胎盘组织碾磨液中氧化指标脂质过氧化氢(LHP)、活性氧(ROS)的含量高于正常对照组产妇,抗氧化指标谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)的含量低于正常对照组产妇,差异有统计学意义(均P<0. 05);子痫前期组产妇胎盘组织碾磨液中炎症指标高迁移率族蛋白(HMGB1)、C反应蛋白(CRP)、白介素-6 (IL-6)的含量高于正常对照组产妇,差异有统计学意义(均P<0. 05)。结论子痫前期产妇胎盘组织中Gas6、SATB1 mRNA表达量存在异常,可客观反映机体氧化应激、炎性反应程度。 Objective To explore the contents of Gas6 and SATB1 in placental tissue of preeclampsia women and their correlations with oxidative stress and inflammatory reaction,provide a reference basis for clinical diagnosis and treatment.Methods Ninety preeclampsia women giving birth to their babies in People's Hospital of Wugong County from July 2014 to February 2017 were collected as preeclampsia group,50 healthy pregnant women giving birth to their babies during the same period were selected as normal control group.Placental tissue samples were collected,then the expression levels of Gas6 and SATB1 mRNA were detected,ELISA was used to detect the levels of oxidative stress and inflammatory reaction indexes in placental tissue fluid.Results The expression level of Gas6 mRNA in placental tissue of preeclampsia group was statistically significantly higher than that of normal control group,the expression level of SATB1 mRNA was statistically significantly lower than that of normal control group(P<0.05);oxidation indexes(LHP,ROS) in placental tissue fluid of preeclampsia group were statistically significantly higher than those of normal control group(P<0.05);antioxidant indexes(GSH-Px,CAT) were statistically significantly lower than those of normal control group(P<0.05);inflammation indexes(HMGB1,CRP,IL-6) in placental tissue fluid of preeclampsia group were statistically significantly higher than those of normal control group(P<0.05).Conclusion The expressions level of Gas6 and SATB1 mRNA in placental tissue of preeclampsia women are abnormal,which can objectively reflect oxidative stress and inflammatory reaction.
