益气健脾通便方对慢传型便秘大鼠结肠组织ICC及SCF/c-kit信号通路的影响 被引量:25

Effect of Yiqi Jianpi Tongbian Recipe on ICC and SCF/c-kit Signaling Pathway in Colon Tissue of Slow Transport Type Constipation Rats

摘要 目的:通过观察益气健脾通便方对慢传输型便秘模型大鼠结肠组织ICC表达的影响及对SCF/c-kit信号通路的修复作用,从理论上揭示益气健脾通便方治疗慢传输型便秘的可能作用机制。方法:采用"复方地芬诺酯灌胃法"复制慢传输型便秘大鼠动物模型,分别运用益气健脾通便方低、中、高剂量干预,并与西药聚乙二醇4000散对比。观察造模及治疗过程中各组大鼠粪便粒数的变化;采用免疫组化法检测大鼠结肠组织ICC的表达量及Western-Blot、RT-PCR法检测大鼠结肠组织c-kit、SCF蛋白的表达及c-kit、SCF mRNA的表达水平。结果:与模型组比较,聚乙二醇组、益气健脾通便方低、中、高剂量组均能明显提高慢传输型便秘大鼠24 h粪便粒数(P<0.05);与模型组比较,聚乙二醇组、益气健脾通便方低、中、高剂量组大鼠结肠组织ICC、c-kit、SCF蛋白及c-kit、SCF mRNA表达水平均显著升高(P<0.05);与聚乙二醇组比较,益气健脾通便方中剂量组大鼠结肠组织ICC及c-kit、SCF蛋白表达均有所升高,但差异无统计学意义(P>0.05);益气健脾通便方中剂量组大鼠结肠组织c-kit、SCF mRNA表达水平较聚乙二醇组明显升高(P<0.05);益气健脾通便方高剂量组大鼠结肠组织c-kit、SCF mRNA表达水平较聚乙二醇组有所升高,但差异无统计学意义(P>0.05)。结论:益气健脾通便方治疗慢传输型便秘疗效显著,其作用机制可能与其提高结肠组织ICC的表达量及修复SCF/c-kit信号通路有关,从而改善便秘症状,以中、高剂量组疗效最为显著。 Objective:Through the observation of Yiqi Jianpi Tongbian Recipe on expression of interstitial cells of Cajal in colon tissue of rats with slow transit constipation model and the repaired effect on SCF/c-kit signal pathway.It theoretically reveals the possible mechanism of Yiqi Jianpi Tongbian Recipe treating slow transit constipation.Methods:We used Compound Diphenoxylate gavage method to replicate animal models of slow transit constipation in rats and used low, medium and high dose interventions of Yiqi Jianpi Tongbian Recipe respectively.We compared western medicine polyethylene glycol 4000 powder.We observed the change of the number of feces pellets in rats of each group during modeling and treatment.The expression of ICC in colon tissue was detected by immunohistochemistry and the expression of c-kit and SCF protein and the expression of c-kit and SCF mRNA in rats colon were detected by Western-Blot and RT-PCR.Results:Compared with the normal group,Polyethylene glycol group and Yiqi Jianpi Tongbian Recipe low, middle and high dose groups all significantly increased the number of fecal pellets in rats with slow transit constipation(P<0.05).Compared with the model group, the expressions of ICC, c-kit and SCF protein, c-kit and SCF mRNA in the colon tissue of rats with polyethylene glycol, Yiqi Jianpi Tongbian recipe low, middle, and high dose groups significantly increased(P<0.05).Compared with the polyethylene glycol group, the expressions of ICC, c-kit and SCF protein in the colon of the middle-dose group of Yiqi Jianpi Tongbian Recipe was increased, but the difference was not statistically significant(P>0.05).The expression of c-kit and SCF mRNA in the colon tissue of the middle-dose group of Yiqi Jianpi Tongbian Recipe was significantly higher than that of the polyethylene glycol group(P<0.05).The expression levels of c-kit and SCF mRNA in the colon tissue of high-dose Yiqi Jianpi Tongbian Recipe group were higher than those in the polyethylene glycol group, but the difference was not statistically significant(P>0.05).Conclusion:The therapeutic effect of Yiqi Jianpi Tongbian Recipe on STC is significant,and its mechanism may be related to increasing the expression of ICC in colon tissue and repair SCF/c-kit signal transduction pathway so as to improve the symptoms of constipation and the most significant effect was seen in the middle and high dose groups.
