多巴丝肼对肺癌A549细胞增殖和凋亡的影响 被引量:3

The Effect of Levodopa and Benserazide Hydrochloride on Lung Cancer A549 Cell Proliferation and Apoptosis

摘要 研究多巴丝肼(含左旋多巴和苄丝肼)对肺癌A549细胞增殖和凋亡的影响。采用含有不同浓度(20,50和100μmol/L)左旋多巴的多巴丝肼处理肺癌A549细胞48 h,应用MTT(噻唑蓝)法测定细胞的增殖活性,再检测乳酸脱氢酶(LDH)活性,Hoechst 33258染色法检测细胞的凋亡情况。结果显示多巴丝肼能以浓度依赖的方式抑制A549细胞的增殖。对照组与处理组之间LDH活性比较均无显著性差异(P> 0. 05)。而50μmol/L多巴丝肼处理48 h能够显著诱导A549细胞凋亡。可见多巴丝肼能够显著抑制肺癌A549细胞增殖,这与其诱导细胞凋亡途径有关。 Chemotherapy is one kind of main therapies to treat lung cancer. While,drug resistance and toxic side effect make the effect and application of chemotherapy limited. So,it is necessary to find out novel effective and safe antitumor drug. It is reported that Levo dopa derivatives have antitumor effect. While,there are no reports about the effect of Levodopa itself or Levodopa and Benserazide Hydrochloride on lung cancer A549 cells. So,this experiment is to find out whether Levodopa and Benserazide Hydrochloride can inhibit lung cancer A549 cells and to explore the inhibition mechanism. The A549 cells have been treated with different concentrations( 20,50 and 100 μmol/L) of Levodopa and Benserazide Hydrochloride for 48 h. Then the proliferative activity was determined by the MTT assay,and the apoptosis was determined by Hoechst 33258 staining assay. Lactate dehydrogenase( LDH) was also tested. The proliferation of the A549 cells was significantly suppressed by the drug in a dose-dependent manner. After A549 cells were treated with Levodopa and Benserazide Hydrochloride at a concentration of 20,50 or 100 μmol/L for 48 h,the proliferative activity decreased from 100% to 91%,64% or 27% respectively. After treated with Levodopa and Benserazide Hydrochloride at the concentration of 50 μmol/L,A549 cells were observed to be decreased and shrunk. However,there were no significant differences( P >0. 05) in the LDH release of the cells between the control group and the drug treatment group. After incubated with 50 μmol/L of Levodopa and Benserazide Hydrochloride for 48 h,shrink and bright A549 cells increase by Hoechst 33258 staining assay. The result indicates that significant apoptosis was induced in A549 cells. Levodopa and Benserazide Hydrochloride could inhibit the proliferation of A549 cells by inducing cellular apoptosis. The drug may be a clue to find effective and safe drug for lung cancer.
