姜黄素通过NF-κB途径诱导鼻咽癌CNE-2Z细胞凋亡

摘要 目的探讨姜黄素对鼻咽癌CNE-2Z细胞的生长抑制作用及诱导凋亡作用并研究其机制。方法MTT法检测姜黄素对鼻咽癌CNE-2Z细胞生长状况的影响;流式细胞仪检测鼻咽癌细胞的凋亡率变化; Western blot检测Bcl-2、Bax、Caspase-3、Fas、以及NF-κB蛋白表达水平变化。Real-time PCR检测caspase家族中caspase-3、caspase-7、caspase-8、caspase-9 mRNA表达水平的变化。结果凋亡率检测结果显示,姜黄素组肿瘤细胞的凋亡率明显高于对照组(P <0. 01)。姜黄素组CNE-2Z细胞的Bcl-2蛋白、NF-κB蛋白的表达水平下降,Bax蛋白、cleaved Caspase-3蛋白的表达水平增高,而Fas蛋白的表达水平未发生明显变化。姜黄素组caspase-3、caspase-7和caspase-9 mRNA表达水平高于对照组。结论姜黄素可以诱导鼻咽癌细胞凋亡,其机制可能是通过抑制Bcl-2基因的表达及NF-κB活性来实现的。 Objective To investigate the effect of curcumin on inhibition of proliferation and induction of apoptosis of nasopharyngeal carcinoma CNE-2Z cells,and to explore the potential mechanisms. Methods The effect of curcumin on nasopharyngeal carcinoma CNE-2Z cells proliferation was assayed by MTT method. After applying curcumin,apoptosis rate changes of nasopharyngeal carcinoma cells were tested by flow cytometry. Bcl-2,Bax,Caspase-3,Fas and NF-κB protein levels were detected by Western blot. Caspase family including caspase-3,caspase-7,caspase-8 and caspase-9 mRNA expression level changes was tested by Real-time PCR. Results The results of apoptosis rate detection showed that curcumin on apoptosis of tumor cells was significantly higher than the control group( P<0.01). Bcl-2 and NF-κB protein expression levels of curcumin group CNE-2Z cell were decreased,and Bax and cleaved Caspase-3 protein expression levels were increased,but the protein expression level of Fas did not change significantly. For curcumin group,the expression levels of caspase-3,caspase-7,and caspase-9 mRNA were higher than those of control group. Conclusion Curcumin can induce apoptosis of nasopharyngeal carcinoma cells,the mechanism of which may be achieved by inhibiting the expression of Bcl-2 gene and the activity of NF-κB.
