摘要
目的探讨缺氧后人脐静脉内皮细胞(HUVECs)可否生成醛固酮(Ald)。方法根据缺氧时间不同,实验分为缺氧0 h(正常组)、4 h、8 h、12 h、24 h组,采用MTT法测定缺氧后细胞活力,ELISA法测定缺氧后醛固酮的产生,RT-PCR法检测盐皮质激素受体(MR) mRNA表达。结果 (1)随着缺氧时间的延长,细胞活性降低;(2)ELISA实验显示,缺氧后伴有醛固酮的产生增加;(3)RT-PCR结果显示,与0 h组比较,缺氧8 h、12 h、24 h后盐皮质激素受体(MR) mRNA表达增加(P <0. 05)。结论在缺氧状态下,HUVECs可以产生Ald,并且在缺氧24 h内其生成量随时间延长而增加。
Objective To investigate whether umbilical vein endothelial cells( HUVECs) can generate aldosterone( Ald) after hypoxia. Methods According to the different time of hypoxia,HUVECs were divided 5 groups,0 h( normal group),4 h,8 h,12 h,24 h group. The survival rate of cells after hypoxia was detected by MTT method,the amount of aldosterone was determinated by ELISA assay,and a real-time fluorescent quantitative assay was utilized to monitor the transcriptional changes in mineralocorticoid receptor( MR). Results(1) With the extension of hypoxia,cell activity decreased;(2) ELISA experiment showed that the generation of aldosterone increased after hypoxia;(3) RT-PCR experiments showed that after hypoxia 8,12,24 hours,the mRNA level of mineralocorticoid receptor( MR) increased( P < 0. 05).Conclusion Under hypoxic conditions,HUVECs can generate Ald,and within 24 hours its generation increased over time.
