小檗碱对PCOS患者卵巢颗粒细胞雄激素分泌及糖代谢的影响 被引量:4

Effects of berberine on androgen secretion and glucose metabolism in ovarian granulosa cells from patients with polycystic ovary syndrome

摘要 目的探讨小檗碱对多囊卵巢综合征(PCOS)卵巢颗粒细胞睾酮分泌及糖代谢异常的改善作用,为小檗碱治疗PCOS的临床研究提供理论依据。方法将15例患者分为:对照组(非PCOS患者5例)、PCOS组(PCOS患者5例)、小檗碱组(PCOS患者5例),三组患者的卵巢颗粒细胞进行体外培养48h后,对小檗碱组卵巢颗粒细胞给予小檗碱干预,其他两组卵巢颗粒细胞不予干预,继续培养48 h。利用放射免疫法检测三组细胞上清液中的睾酮(T)的水平;利用葡萄糖测定试剂盒(HK法)检测三组细胞培养液中的残糖量;利用2-NBDG检测三组细胞的葡萄糖摄取情况。结果与对照组相比,PCOS组T水平增高(P <0. 05),培养液残糖量增加(P <0. 01),2-NBDG摄取量降低(P <0. 01);与PCOS组相比,小檗碱组T水平降低(P <0. 05),培养液残糖量减少(P <0. 01),2-NBDG摄取量增加(P <0. 01);小檗碱组与对照相比,T水平、培养液残糖量、2-NBDG摄取量均无显著差异(P> 0. 05)。结论 PCOS患者卵巢颗粒细胞雄激素分泌增加且存在糖代谢障碍,小檗碱能够改善其糖代谢异常并能抑制雄激素的分泌。 Objective To explore the effect of berberine on the secretion of testosterone and abnormal glucose metabolism in ovarian granulosa cells of polycystic ovary syndrome( PCOS),and to provide a theoretical basis for clinical research of berberine in the treatment of PCOS.Methods Fifteen cases were divided into three groups: the control group( 5 cases of non PCOS),PCOS group( 5 cases of PCOS),and berberine group( 5 cases of PCOS),then were cultured in vitro for 48 h. Then berberine was used to intervene the granulosa cells in berberine group,the other two groups were not intervened,and cultured for 48 hours. The level of testosterone was determined by radioimmunoassay;cell culture medium residual sugar was tested by utilizing glucose assay kit( HK method);glucose uptake in granulosa cells was detected by 2-NBDG as fluorescence probe. Results Compared with the control group,testosterone secretion within ovarian granulosa cells from PCOS group was increased( P < 0. 05),the residual sugar in the culture medium was increased( P < 0. 01),but 2-NBDG uptake in granulosa cells was decreased( P < 0. 01). The level of testosterone( P < 0. 01) and residual sugar( P <0. 05) in berberine group was significantly decreased,which compaired with PCOS group,2-NBDG showed glucose uptake in berberine group was increased( P < 0. 01). There was no significant difference in T level,residual sugar and 2-NBDG uptake between the berberine group and control group( P > 0. 05). Conclusion There is an increase in testosterone secretion and impaired glucose metabolism in granulosa cells of PCOS patients. Berberine can improve the abnormal glucose metabolism and reduce testosterone secretion.
