藏红花素对失血性休克所致器官损伤保护作用 被引量:4

Protective Effects of Crocin on Organ Damage in Hemorrhagic Shock Rats

摘要 目的研究天然抗氧化剂藏红花素(crocin)对失血性休克(hemorrhagic shock, HS)大鼠氧化应激损伤和器官组织损伤的保护作用。方法 80只实验用Wistar大鼠采用数字表法随机分为假手术组(空白组)、失血性休克组(模型组)、失血性休克+生理盐水(对照组)、失血性休克+藏红花素(实验组),每组20只,采用颈动脉放血的方法制备失血性休克大鼠模型;开始复苏时静脉注射给药。记录给药前和给药24h后平均动脉压(MAP)、心率(HR)并测定血液pH值、PO_2和pCO_2;记录给药24h后的存活率;称量并计算肺组织湿/干比重(W/D),HE染色后测定肺组织定量评估指数(IQA);测定血清中血尿素氮(BUN)、肌酐(creatinine)、丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶氨酶(AST)、丙二醛(MDA)、谷胱甘肽(GSH)含量,测定肺组织中丙二醛(MDA)、谷胱甘肽(GSH)含量,测定肝脏、肾脏、肺组织中髓过氧化物酶(MPO)活性。结果给药24h后,实验组HR、pH值、pO_2显著升高而pCO_2显著降低(P <0.05),MAP比较差异无统计学意义(P>0.05);存活率显著升高(P<0.01);肺组织W/D显著降低(P<0.05)、IQA显著降低(P<0.01);血清中BUN、Creatinine、ALT、AST、MDA含量显著降低且GSH含量显著升高(P <0.05);肺组织MDA含量显著降低且GSH含量显著升高(P<0. 05);肝脏、肾脏、肺组织中MPO活性均显著降低(P<0.05)。结论藏红花素能够抑制HS引起的肝脏、肾脏、肺组织损伤,其作用机制可能与藏红花素能够减轻氧化应激损伤有关。 Objective To study the protective effects of natural antioxidants crocin on oxidative stress and organ damage in hemorrhagic shock(HS) rats. Methods Eighty rats were randomly devided into four groups: sham(blank group), HS(model group), HS +NS(control group), HS + Crocin group(test group), n = 20 HS rat models were prepared by carotid artery bleeding, and the drugs were given intravenously at the beginning of resuscitation. The MAP, HR were recorded before and 24 h after resuscitation. The pH, p02,pCO2 of blood were determined also. The W/D ratio of lung tissue was calculated and the IQA was calculated after HE staining. The content of BUN, ALT, AST, MDA, GSH in serum and the content of MDA, GSH were determined by ELISA. The activity of MPO in lung,renal and hepatic tissue were determined. Results Compared with control group, before the drugs were given, the difference of measured indexes in test group had no statistical significance(P >0.05). 24 h after the drugs were given, the HR,pH,p02 in test group were significantly increased and the p02 was significantly decreased(P <0.05), and the difference of MAP between two group had no statistical significance(P>0.05);the survival rate increased significantly(P<0.01). The W/D ratio and IQA of lung tissue were significantly decreased(P<0.05 orP<0.01). The content of BUN, Creatinine, ALT, AST, MDA in serum were significantly decreased and GSH was significantly increased(P<0.05 orP<0.01). The content of MDA in lung tissue was significantly decreased and GSH was significantly increased(P < 0.05). The activity of MPO in lung, renal and hepatic tissue were significantly decreased(P <0.05 or P<0.01). Conclusion Crocetin can reduce HS-induced liver,kidney a d lung injury, which perhaps related to its effects of inhibiting oxidative stress injury.
