摘要
目的研究倒吊笔根的化学成分,并初步评价其单体的抗肿瘤活性。方法采用硅胶柱色谱、Sephadex LH-20凝胶柱色谱、制备高效液相色谱及重结晶等方法进行分离纯化,通过波谱数据和理化性质进行结构解析、鉴定。MTT法检测化合物体外抗肿瘤活性。结果从倒吊笔根75%乙醇提取物三氯甲烷部位中分离鉴定了16种化合物,分别鉴定为:尿嘧啶(uracil,1),尿囊素(allantoin,2),喹唑啉-2,4(1H,3H)-二酮(2,4(1H,3H)-quinazolinedione,3),色胺酮(tryptanthrin,4),槲皮素(quercetin,5),染料木苷(genistin,6),蒙花苷(linarin,7),豆甾醇(3β-hydroxysitost-5-en-7-one,8),5,6α-epoxy-5α-stigmastan-3α-ol(9),5,6β-epoxy-5β-stigmastan-3β-ol(10),桦木酸(betulinic acid,11),桦木酮酸(betulonic acid,12),齐墩果酸(oleanolic acid,13),3β-乙酰基-12-烯-11-酮(3β-acetoxy-urs-12-ene-11-one,14),3β,23-dihydroxyolen-12-en-28-oic acid(15),N-(2'-hydroxyeicoxyicosanoyl)-1,3,4-trihydroxy-2-aminoheptadec-5-ene(16)。化合物13对人乳癌细胞株MCF-7、人肝癌细胞株Hep G2、人结肠癌细胞株HCT116、人宫颈癌细胞株He La的生长有一定的抑制作用,其IC50值分别为8. 6、9. 2、13. 8、12. 3μmol/L。结论化合物1~10,13,15,16为首次从该植物中分离得到。化合物4,11,12,13在体外具有抑制肿瘤细胞生长的活性。
Objective To investigate the chemical constituents of roots from Wrightia pubescens. Methods Compounds were isolated by the combined use of column chromatography,preparative HPLC and recrystallization,and their structures were identified by their physicochemical and spectroscopic data. Antitumor activities of the compounds were evaluated using MTT method in vitro. Results Sixteen compounds 1-16 were isolated from the trichloromethane soluble part of 75%ethanol extract of the root of W. pubescens,and identified as uracil( 1),allantoin( 2),2,4( 1 H,3 H)-quinazolinedione( 3),tryptanthrin( 4),quercetin( 5),genistin( 6),linarin( 7),3β-hydroxysitost-5-en-7-one( 8),5,6α-epoxy-5α-stigmastan-3α-ol( 9),5,6β-epoxy-5β-stigmastan-3β-ol( 10),betulinic acid( 11),betulonic acid( 12),oleanolic acid( 13), 3β-acetoxy-urs-12-ene-11-one( 14), 3β, 23-dihydroxyolen-12-en-28-oic acid( 15), N-( 2’-hydroxyeicoxyicosanoyl)-1,3,4-trihydroxy-2-aminoheptadec-5-ene( 16). Compound 13 showed antitumor activity against Hep G2,HCT116,He La and MCF-7 cells in vitro with IC50 of 8. 6,9. 2,13. 8 and 12. 3 μmol/L,respectively.Conclusion Compounds 1-10,13,15,and 16 are isolated from the genus Wrightia for the first time. Compounds 4,11,12,13 show cytotoxic activities against tumor cells in vitro.
