胃癌患者microRNA-126水平与其放化综合疗效的关系及对其胃癌细胞放射增敏作用的影响

Relationship between microRNA-126 level in gastric cancer patients and comprehensive radiotherapy efficacy and the effect on radiosensitization of gastric cancer cells

目的探究胃癌患者microRNA-126水平与其放化综合疗效的关系及对其胃癌细胞放射增敏作用的影响。方法回顾性选取2015年4月至2016年10月商洛市中心医院肿瘤诊治中心收治的75例晚期胃癌患者作为研究对象。观察临床资料、放化疗效果不同的患者中MicroRNA-126相对表达水平,以及转染情况和放射增敏作用。结果中、高分化组患者的microRNA-126相对表达水平为(1.14±1.40)2-△△Ct,明显低于低分化的(1.97±2.05)2-△△Ct,差异有统计学意义(P<0.05);Ⅲ～Ⅳ期患者的microRNA-126相对表达水平为(1.02±1.27)2-△△Ct,明显低于Ⅰ～Ⅱ期的(1.84±1.97)2-△△Ct,差异有统计学意义(P<0.05);直径<5 cm组患者的microRNA-126相对表达水平为(2.17±2.24)2-△△Ct,明显高于直径≥5 cm的(1.16±1.16)2-△△Ct,差异具有统计学意义(P<0.05);非敏感组患者的组织、血浆的microRNA-126相对表达水平分别为(0.73±0.07)2-△△Ct、(0.57±0.03)2-△△Ct,显著低于敏感组的(1.00±0)2-△△Ct、(1.00±0)2-△△Ct,差异具有统计学意义(P<0.01);转染microRNA-126组患者的D0、SF分别为(2.35±0.29)、(0.53±0.15),明显低于未转染组、转染microRNA-NC组的(4.12±0.25)、(0.84±0.06),(4.24±0.24)、(0.79±0.09),而SER为(1.79±0.1),明显高于其他两组的(1.00±0)、(1.06±0.11),差异均具有统计学意义(P<0.05);未转染组与转染microRNA-NC组的D0、SF、SER比较差异均无统计学意义(P>0.05)。结论 microRNA-126可能具有放射增敏作用,并提高放化疗效果;且在不同胃癌患者中microRNA-126相对表达水平有差异,可能提示其具有诊断胃癌的作用。

Objective To explore the relationship between micro RNA-126 level in gastric cancer patients and comprehensive radiotherapy efficacy and the effect on radiosensitization of gastric cancer cells. Methods A total of 75 patients with advanced gastric cancer were retrospectively selected as the research subjects, who were admitted to the Cancer Diagnosis and Treatment Center at Shangluo Central Hospital from April 2015 to October 2016. Clinical data of these patients were observed, as well as the relative expression level of micro RNA-126 in patients with different chemoradiotherapy effects, transfection, and radiosensitization. Results The relative expression level of micro RNA-126 in patients in the moderate to high differentiation group was(1.14±1.40)2-△△Ct, which was significantly lower than(1.97±2.05)2-△△Ctin the low differentiation group, and the difference was statistically significant(P<0.05). The relative expression level of microRNA-126 in stage Ⅲ-Ⅳ patients was(1.02±1.27)2-△△ Ct, which was significantly lower than(1.84±1.97)2-△△ Ctin stage Ⅰ-Ⅱ(P<0.05). The relative expression level of microrna-126 in the group with a tumor diameter <5 cm was(2.17±2.24)2-△△ Ct, which was significantly higher than(1.16±1.16)2-△△ Ctin the group with a tumor diameter ≥5 cm(P<0.05). The relative expression levels of microRNA-126 in tissues and plasma of non-sensitive group were respectively(0.73±0.07)2-△△Ctand(0.57±0.03)2-△△Ct, which were significantly lower than(1.00±0)2-△△Ctand(1.00±0)2-△△Ctin the sensitive group, respectively(P<0.01). The D0 and SF of the transfected microRNA-126 group were respectively(2.35±0.29)and(0.53 ± 0.15), which were significantly lower than(4.12 ± 0.25) and(0.84 ± 0.06) in the non-transfected group and(4.24±0.24) and(0.79±0.09) in the transfected microRNA-NC group;and the SER was(1.79±0.1), which was significantly higher than(1.00±0) and(1.06±0.11), respectively, in the other two groups(P<0.05). There were no statistically significant differences in D0, SF and SER between non-transfected group and transfected microRNA-NC group(P>0.05). Conclusion MicroRNA-126 may have radiosensitizing effects and can improve the effect of radiotherapy and chemotherapy. In addition, the relative expression level of microRNA-126 is different in different gastric cancer patients, which may indicate its function in the diagnosis of gastric cancer.
