不同基因型兰花组培苗对植物生长调节剂的响应 被引量:9

The Response of Tissue Culture Seedlings of Different Genotypes Orchids to Plant Growth Regulator

摘要 组织培养技术是兰花苗实现工厂化标准化生产的重要技术手段之一,利用植物生长调节剂可改善兰花组培苗品质、提高出瓶存活率。本研究以3个原生种和5个杂交F1代兰花种子为组培材料,设置4种处理(CK, 0.1 mg/L TDZ, 0.5 mg/L PP333, 0.1 mg/L TDZ+0.5 mg/L PP333),研究植物生长调节剂及组合对不同基因型兰花组培苗株型的影响,其中株高及根长做为兰花试管苗的主要株型特征。结果表明,植物生长调节剂处理与对照CK组相比显著降低了8个基因型兰花的株高,抑制根的伸长,促进茎与根的横向生长;0.1 mg/L TDZ处理下兰花品种株高与对照CK组无显著差异,但根长与对照存在极显著差异,而0.5 mg/L PP333,0.1 mg/L TDZ+0.5 mg/L PP333处理后株高及根长均缩短,与对照差异均显著;不同基因型兰花的生长对同一植物生长调节剂敏感性存在极显著差异(p<0.01);根据兰花组培苗对植物生长调节剂的响应情况,可分为3类:TDZ敏感型、PP333敏感型、TDZ+PP333敏感型。 The technology of tissue culture is one of the important technical means to realize standardized production of orchids seedlings. The use of plant growth regulator can improve the quality of orchids tissue culture seedlings and the survival rate after they come out of bottles. In this study, we chose three kinds of primary seeds and five F1 hybrids seeds of orchids as tissue culture planting materials, and designed 4 kinds of treatments(CK,0.1 mg/L TDZ, 0.5 mg/L PP333 and 0.1 mg/L TDZ+0.5 mg/L PP333). The experiment was used to study the effects of plant growth regulators and combinations on the plant-type of different genotypes orchids tissue culture seedlings. The plantlet height and root length were recorded as the main plant-type characteristics of the plantlets.The results showed that compared with control CK group, the plant height of 8 genotypes of orchids was significantly reduced, the elongation of root was inhibited, and the lateral growth of stem and root was promoted through plant growth regulator treatment;The plantlet height of orchids species under 0.1 mg/L TDZ treatment had no significant difference compared with control CK group, but the root length had extremely significant difference compared with CK group, while the plant height and root length were shortened after the treatment of 0.5 mg/L PP333 and 0.1 mg/L TDZ +0.5 mg/L PP333, which had significant difference than control group. The result indicated that the sensitivity of different orchids genotypes to plant growth regulator had significant difference(p<0.01).According to the response of orchids tissue culture seedlings to plant growth regulators, they could be divided into three categories: TDZ-sensitive, PP333-sensitive and TDZ+PP333-sensitive.
