复方辛夷滴鼻液对变应性鼻炎大鼠外周血中T-betmRNA、GATA-3mRNA、OX40LmRNA表达量的影响 被引量:1

摘要 目的:研究复方辛夷滴鼻液对变应性鼻炎大鼠外周血中T-betmRNA、GATA-3mRNA、OX40LmRNA表达量的影响。方法卵清蛋白(致敏原)+氢氧化铝(佐剂),制成OVA混悬液。将60只大鼠随机分为AR组(A组)、AR+复方辛夷滴鼻液治疗组(B组)、健康空白对照组(C组),每组20只。于基础致敏和局部激发后,观察大鼠行为,并记录症状体征积分,应用PCR法检测大鼠外周血中T-betmRNA、GATA-3mRNA、OX40LmRNA的表达。结果观察并记录大鼠症状体征,A组与C组比较,差异具有统计学意义(P<0.05),表明AR大鼠造模成功。三组大鼠外周血中T-betmRNA的表达量A组(1.01±0.16)、B组(1.32±0.17)、C组(1.79±0.29),三组两两比较,差异有统计学意义(P<0.01);GATA-3mRNA的表达量:A组(1.01±0.16)、B组(0.43±0.05)、C组(0.31±0.04),B组与A、C组比较差异有统计学意义(P<0.01);OX40LmRNA的表达量为A组(1.01±0.17)、B组(0.35±0.04)、C组(0.30±0.05),A组与B组,A组与C组比较,P<0.01,差异有统计学意义。而B组与C组比较,P=0.051>0.05,差异无统计学意义。结论复方辛夷滴鼻液能够明显改善AR大鼠的过敏症状与体征,并且可以降低变应性鼻炎大鼠外周血中GATA-3mRNA和OX40LmRNA的相对表达量、升高外周血中T-betmRNA的表达量。 Objective:To study the effect of compound Xinyi nasal drops on the expression of T-bet,GATA-3 and OX40L in peripheral blood of rats with allergic rhinitis.Methods:Ovalbumin (Sensitinogen)±aluminum hydroxide (Adjuvant)was used to make OVA suspension.Sixty rats were randomly divided into AR group (group A),AR ±compound Xinyi nasal drops treatment group (group B)and healthy blank control group (group C),20 rats in each group. After basic sensitization and local stimulation,the behavior of rats was observed,and the scores of symptoms and signs were recorded.The expressions of T-bet,GATA-3 and OX40L in peripheral blood of rats were detected by PCR.Results:The symptoms and signs of AR rats were observed and recorded.There was a significant difference between group A and group C (P<0.05),which indicated that AR rats were successful in modeling.The expression of T-bet in peripheral blood of rats in three groups was (1.01±0.16),(1.32±0.17)and (1.79±0.29),respectively.There was sig- nificant difference between the three groups (P<0.01).The expression of GATA-3 was (1.01±0.16),(0.43±0.05)and (0.31 ±0.04)in group A,B and C,respectively.The expression of OX40L in group A (1.01±0.17),group B (0.35±0.04), group C (0.30±0.05),group A compared with group B,group A compared with group C,the difference was statistically significant (P<0.01).There was no significant difference in P=0.051>0.05 between group B and group C.Conclusion: Compound Xinyi Nasal Drops can significantly improve the allergic symptoms and signs of AR rats,and can reduce the relative expression of GATA-3 and OX40L in peripheral blood of rats with allergic rhinitis,and.increase the expression of T-bet in peripheral blood.
