诃子细粉混悬液对清醒Beagle犬心血管系统及体温的影响

摘要 目的观察单次灌胃给予诃子细粉混悬液后对清醒Beagle犬心血管系统及体温的影响。方法设溶媒对照组(0.5%CMC-Na)及诃子细粉混悬液低、中、高剂量组(0.08、0.36、1.6 g·kg^(-1)),选用6只Beagle犬,设4个给药周期,每两个给药周期间设7 d的洗脱期;使用植入式生理信号遥测系统(DSI),采集给药前1~2h及给药后24h清醒Beagle犬的心电、血压及体温参数。结果与溶媒对照组比较,诃子细粉混悬液低、中、高剂量组清醒Beagle犬心电、血压参数均未出现明显改变;低剂量组在给药后14h采集点体温出现具有统计学意义的降低(P <0.05),但与给药前比较差异无统计学意义,随后恢复正常;诃子细粉混悬液中、高剂量组与溶媒对照组比较差异无统计学意义。结论本实验条件下,诃子细粉混悬液在0.08~1.6 g·kg^(-1)给药剂量内对清醒Beagle犬心电、血压及体温无明显影响。 Objective To determine the effect of single oral administration of Chebulae fructus suspension on the cardiovascular system and body temperature in conscious Beagle dogs. Methods Four dose groups were designed in this study: a vehicle control group(0.5%CMC-Na), a low dose, a medium dose and a high dose Chebulae fructussuspension group(0.08, 0.36, and 1.6 g·kg^-1). Six Beagle dogs were selected for the four dosing cycles. A washout period of 7 days was set for every two administration weeks. DSI implantable physiological signal telemetry system was applied to acquire the ECG, blood pressure, and body temperature. Results Compared with the vehiclel control group, there was no significant difference in ECG and blood pressure parameters between the low medium and high dose Chebulae fructus suspension groups of conscious Beagle dogs. The low dose of Chebulae fructus suspension group showed a significant decrease in the body temperature at the post-collection point after administration for 14 hours, but there was no significant difference as compared with that of pre-dose and then returned to normal. No significant difference was found between the medium and high dose group with the control group. Conclusion Under the experiment conditions, no obvious effect on the electrocardiogram, blood pressure and body temperature in the conscious Beagle dogs has been found within the dose range of 0.08-1.6 g·kg^-1.
