摘要
提出基于荧光定量分析方法对离体AGEs进行定量检测的新方法。将AGEs标准品溶液配成7种不同浓度,通过荧光分光光度计进行荧光光谱检测,取荧光峰值强度和浓度作标准工作曲线,再配比以上7种浓度之外的溶液对标准工作曲线的准确性进行检验。研究发现选用370 nm作为AGEs激发波长,可观察到荧光光谱在470 nm处存在明显峰值,且随着浓度的增大,荧光峰值也随之增大。标准工作曲线的相关系数R^2=0. 997。通过标准工作曲线测得的浓度值与实际浓度值的相关系数R^2=0. 997。实验结果表明,荧光定量检测方法在离体AGEs检测中具有可行性。
A new method for quantitative detection of AGEs in vitro based on fluorescence quantitative analysis was proposed.The AGEs standard solution was diluted into 7 different concentrations,and then the fluorescence spectrophotometer was used to detect the fluorescence spectrum of AGEs.The peak intensity of fluorescence and concentration of AGEs were used to make standard working curves.The accuracy of the standard working curve was checked by detecting other solution with the concentrations beyond the above 7 concentrations.It was found that the fluorescence spectrum had obvious peak at 470 nm when 370 nm was used as the excitation wavelength of AGEs.With the increase of concentration,the fluorescence peak also increased.The correlation coefficient of the standard working curve was 0.997.The correlation coefficient between the concentration measured by the standard working curve and the actual concentration was 0.997.The experimental results showed that fluorescence quantitative detection was feasible in detecting AGEs in vitro.
