丹皮酚注射液对急性胰腺炎模型大鼠血清炎性因子的影响 被引量:12

Effect of Paeonol Injection on Serum Inflammatory Factors in Rats with Acute Pancreatitis

摘要 目的:探究丹皮酚注射液对急性胰腺炎模型大鼠血清TNF-α、IL-1β、IL-6和IL-8的影响。方法:将60只健康的Wistar大鼠随机分为假手术组、模型组、乌司他丁组以及丹皮酚注射液高、中、低剂量组,每组10只。采用5%牛磺胆酸钠溶液逆行注入胰胆管制作急性胰腺炎大鼠模型,模型组和假手术组均在造模前30 min,于大鼠后大腿外侧肌肉注射1.5 mL/kg的生理盐水,乌司他丁组在造模完成之后,立即腹腔注射乌司他丁20,000 U/kg;丹皮酚注射液高、中、低剂量组在造模前30 min于大鼠后大腿外侧肌肉注射丹皮酚注射液(剂量分别为25、15、10mg/kg)。各组大鼠在制作模型6 h后,行腹腔主动脉采血,然后处死大鼠,取大鼠胰腺标本,制成病理切片,并用ELISA法检测大鼠血清TNF-α、IL-1β、IL-6、IL-8的浓度。结果:模型组大鼠胰腺组织病理学评分、血清TNF-α、IL-1β、IL-6及IL-8水平均明显高于假手术组(P<0.01),乌司他丁组和丹皮酚注射液组的上述指标较模型组明显改善,并呈一定的量效关系(P<0.05或P<0.01)。结论:丹皮酚注射液能显著降低急性胰腺炎大鼠血清TNF-α、IL-1β、IL-6、IL-8水平,从而减轻急性胰腺炎的病理损害程度。 Objective: To observe the effect of Paeonol injection on serum tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β), interleukin-6(IL-6) and interleukin-8(IL-8) in rats with acute pancreatitis. Methods: The60 healthy Wistar rats were randomly divided into 6 groups, 10 in each. The 6 groups were Sham group, model group, ulinastatin group, high, mid, and low-dose Paeonol injection groups. Acute pancreatitis model was developed by retrograde injection of 5% sodium taurocholate into pancreatic duct in rats. Model group and sham group were intramuscularly injected 1.5 mL/kg saline on the lateral side of the rat’s posterior thigh 30 min before modeling. Ulinastatin group was injected into the abdomen with ulinastatin at 20,000 U/kg after the modeling was completed. High, mid, and low-dose Paeonol injection groups were intramuscularly injected with Paeonol injection(doses were 25, 15 and 10 mg/kg respectively) on the lateral side of the rat’s posterior thigh 30 min before modeling. The abdominal aortic blood was taken 6 h after modeling and then these rats were put to death. Pancreatic specimens were taken from these rats for making pathological sections. The levels of rat serum TNF-α, IL-1β, IL-6 and IL-8 were detected by enzyme linked immunosorbent assay(ELISA). Results: Compared with sham group, pancreatic histopathological scores and the levels of serum TNF-α, IL-1β, IL-6, IL-8 were significantly increased in model group(P<0.01). The above indexes in ulinastatin group and paeonol injection group were significantly improved compared with model group, and showed a dose-effect relationship(P<0.05 or P<0.01). Conclusions: Paeonol injection can significantly decrease TNF-α, IL-1β, IL-6 and IL-8, and therefore can alleviate pancreatic damage in pancreas in rats with acute pancreatitis.
