针刺人迎穴对自发性高血压大鼠血压及延髓头端腹外侧区氧化应激反应的影响 被引量:13

Effect of Acupuncture at Renying Point(ST9) on Blood Pressure and Oxidative Stress Response of Ventrolateral Medulla Head in Spontaneously Hypertensive Rats

摘要 目的:研究针刺人迎穴对自发性高血压大鼠血压及延髓头端腹外侧区(RVLM)氧化酶丙二醛(MDA)、抗氧化酶超氧化物歧化酶(SOD)的蛋白表达和外周血清丙二醛(MDA)、超氧化物歧化酶(SOD)及去甲肾上腺素(NE)含量的影响。方法:将16周龄雄性自发性高血压大鼠(SHR)30只随机分为3组:模型组、针刺组及电针组,每组10只,以10只16周龄雄性京都Wsitar大鼠(WKY)作为空白组。电针组和针刺组连续针刺人迎穴28 d,1次/d;模型组和空白组进行与实验组同等时间、同等程度的抓取。监测干预前后大鼠的收缩压与舒张压,并计算平均动脉压(MAP)。实验结束后,用蛋白质印迹法(Western Blot)测定RVLM的丙二醛(MDA)、超氧化物歧化酶(SOD)蛋白表达;用酶联免疫吸附(ELISA)测定血清丙二醇MDA、超氧化物歧化酶(SOD)及去甲肾上腺素(NE)含量。结果:与空白组比较,模型组平均动脉压MAP明显升高(P<0.05);针刺干预后,针刺组、电针组MAP显著低于模型组(P<0.05)。与空白组比较,模型组RVLM的MDA蛋白表达水平与血清含量显著升高(P<0.05),模型组RVLM的SOD蛋白表达水平与血清含量显著降低(P<0.05),同时模型组血清NE含量明显升高(P<0.05)。与模型组比较,针刺组与电针组RVLM的MDA蛋白表达水平与血清含量下降(P<0.05),针刺组与电针组RVLM的SOD蛋白表达与血清含量升高(P<0.05),针刺组与电针组血清NE含量明显降低(P<0.05)。结论:针刺人迎穴可以通过提高RVLM抗氧化能力降低交感神经活性从而缓解自发性高血压大鼠高血压反应。 Objective: To study the effects of acupuncture at Renying acupoint(ST9) on the expression of oxidase malondialdehyde(MDA), antioxidant enzyme superoxide dismutase(SOD) in the rostral ventrolateral medulla(RVLM) and the content of malondialdehyde(MDA), superoxide dismutase(SOD) and norepinephrine(NE)in peripheral serum of spontaneously hypertensive rats. Methods: Thirty male 16-week-old spontaneously hypertensive rats(SHR) were randomly divided into three groups: model group, acupuncture group and electroacupuncture group, 10 rats in each group. Male Kyoto Wsitar rats(WKY) served as a blank control group.Electroacupuncture group and acupuncture group continuously intervened for 28 days, acupuncture noce a day;model group and blank control group were the same time as the experimental group, the same degree of grasping. Systolic and diastolic blood pressure were measured before and after intervention in rats, and mean arterial pressure(MAP) was calculated. After the experiment, MDA and SOD protein expression in RVLM were determined by Western Blot. Serum levels of MDA, SOD and NE were measured by enzyme linked immunosorbent assay(ELISA). Results: Compared with the blank group, the mean arterial pressure MAP in the model group increased significantly(P <0.05). After the acupuncture intervention, the MAP in the acupuncture group and the electroacupuncture group was significantly lower than that in the model group(P<0.05). Compared with the blank group, the MDA protein expression level and serum level of RVLM in the model group increased significantly(P<0.05). The SOD protein expression level and serum level of RVLM in the model group decreased significantly(P<0.05). At the same time, serum NE levels in the model group increased significantly(P<0.05).Compared with the model group, MDA protein expression and serum levels of RVLM in acupuncture group and electroacupuncture group were decreased(P <0.05). SOD protein expression and serum level of RVLM in acupuncture group and electroacupuncture group increased(P<0.05), serum NE content in acupuncture group and electroacupuncture group was significantly decreased(P <0.05). Conclusion: Acupuncturing Renying point can relieve sympathetic nerve activity by increasing the antioxidant capacity of RVLM and relieve the hypertensive response.
