摘要
目的探讨骨肉瘤组织环状RNA circ_0002052表达及与临床病理特征和预后的关系。方法骨肉瘤患者46例,取手术切除骨肉瘤组织及癌旁正常组织,采用实时荧光定量PCR法检测环状RNA circ_0002052相对表达量,并分析其表达与临床病理特征的关系;随访3a,Kaplan-Meier生存法分析环状RNA circ_0002052低表达及高表达骨肉瘤患者生存期;多因素Cox回归分析骨肉瘤患者预后不良的影响因素。结果骨肉瘤组织环状RNA circ_0002052相对表达量(0.42±0.02)低于癌旁正常组织(1.04±0.05)(P<0.05);环状RNA circ_0002052高表达骨肉瘤患者无远处转移比率(76.47%)、TNM分期Ⅰ~Ⅱ期比率(70.00%)高于低表达者(23.53%、30.00%)(P<0.05),其表达在年龄、性别比例、肿瘤直径、肿瘤位置、分化程度上差异无统计学意义(P>0.05);环状RNA circ_0002052低表达骨肉瘤患者总生存期[(15.77±0.70)个月]较高表达患者[(27.87±1.17)个月]短(P<0.05);环状RNA circ_0002052低表达(HR=3.298,95%CI:1.531~5.515,P=0.006)是骨肉瘤患者预后不良的危险因素。结论骨肉瘤组织环状RNA circ_0002052表达明显下调,其低表达提示预后不良。
Objective To investigate the expression of circular RNA circ0002052 in osteosarcoma and its correlation with clinicopathological features and prognosis of osteosarcoma.Methods Real-time fluorescence quantitative PCR was performed to detect the expression levels of circular RNA circ0002052 in osteosarcoma tissues and adjacent normal tissues from 46 patients with osteosarcoma.The relationship between circular RNA circ0002052 and clinicopathological features was analyzed.Kaplan-Meier analysis was performed to identify the survival in osteosarcoma patients with lowly and highly expressed circular RNA circ0002052.Multivariate Cox proportional hazard regression method was used to evaluate the influencing factors for poor prognosis of osteosarcoma patients.Results The relative expression of human circular RNA circ0002052 in osteosarcoma tissues(0.42±0.02)was significantly lower than that in adjacent normal tissues(1.04±0.05)(P<0.05).The no-distant metastasis rate and ratio of TNM stageⅠ-Ⅱ were significantly higher in patients with highly expressed circular RNA circ0002052(76.47%,70.00%)than those in patients with lowly expressed circular RNA circ0002052(23.53%,30.00%)(P<0.05),and circular RNA circ0002052 showed no significant differences in the age,gender,tumor diameter,tumor position and differentiating degree(P>0.05).The survival time was significantly shorter in patients with lowly expressed circular RNA circ0002052((15.77±0.70)months)than that in patients with highly expressed circular RNA circ0002052((27.87±1.17)months)(P<0.05).The low expression of circular RNA circ0002052 was the risk factor for poor prognosis in patients with osteosarcoma(HR=3.298,95%CI:1.531-5.515,P=0.006).Conclusion Circular RNA circ0002052 is significantly downregulated in osteosarcoma tissues,indicating apoor prognosis.
