人子宫内膜细胞基因表达谱研究

摘要 目的通过研究人子宫内膜细胞基因表达谱,分析人子宫内膜特异表达基因的水平及相关信号通路。方法通过新一代测序数字基因表达谱(DGE)方法对人子宫内膜细胞进行转录组测序,与人脐带间充质干细胞基因表达谱对比获得子宫内膜特异表达基因,进行GO注释富集分析和KEGG信号通路富集分析。结果人子宫内膜细胞表达基因17 485个。其中与人脐带间充质干细胞基因表达谱对比,子宫内膜细胞特异表达基因3 651个。选取转录本定量FPKM值大于10个拷贝的627个子宫内膜特异表达基因,进行GO注释富集分析发现,子宫内膜特异表达基因与细胞外区域的结构和细胞外基质、胶原蛋白代谢、细胞迁移的调控等功能相关; KEGG信号通路富集分析发现,子宫内膜特异表达基因与黏着斑、吞噬泡、脂肪酸代谢、细胞凋亡等信号通路相关。结论获取人原代子宫内膜细胞的基因表达谱,并对其差异基因进行GO注释和KEGG信号通路富集分析,可为进一步研究提高子宫内膜容受性的方法提供可靠依据。 Objective To analyze the level of human endometrial specific expression genes and the related signal pathways by studying gene expression profile of human endometrial cells. Methods Transcriptome sequencing was performed among human endometrial cells by the new generation sequencing digital gene expression (DGE) profile,endometrial specific genes were obtained by comparing with human umbilical cord mesenchymal stem cells,GO annotation enrichment analysis and KEGG signaling pathway enrichment analysis were carried out. Results There were 17 485 genes expressed in human endometrial cells. Compared with human umbilical cord mesenchymal stem cells,there were 3 651 genes specifically expressed in endometrial cells. A total of 627 endometrial specific genes with FPKM more than 10 copies were selected. GO annotation enrichment analysis showed that endometrial specific genes were correlated with extracellular region part and extracellular matrix,collagen metabolism,regulation of cell migration. KEGG signaling pathway enrichment analysis showed that endometrial specific genes were correlated with focal adhesion,phagosome,fatty acid metabolism,and cell apoptosis. Conclusion Obtaining gene expression profile of human primary endometrial cells,GO annotation enrichment analysis,and KEGG signaling pathway enrichment analysis can provide a reliable basis for further improving human endometrial receptivity.
