棉花蔗糖转化酶基因家族的生物信息学分析 被引量:7

摘要 为更好地了解蔗糖转化酶在棉花基因组中的数量和分布情况等,本研究利用拟南芥中17个蔗糖转化酶家族的基因为参考序列,在海岛棉、陆地棉、雷蒙德氏棉和亚洲棉基因组数据库中鉴定到的蔗糖转化酶基因家族成员分别为65、46、26和25个,结果表明,海岛棉所含有的蔗糖转化酶基因明显多于陆地棉,而雷蒙德氏棉比亚洲棉仅多了1个。通过基因结构分析发现,酸性蔗糖转化酶和中性/碱性蔗糖转化酶α亚族大都包含6或7个外显子,而β亚族大多含有4或5个外显子。此外,部分海岛棉与雷蒙德氏棉在基因结构上存在着较长的UTR区,而陆地棉与亚洲棉可能没有;酸性蔗糖转化酶家族基因所编码的氨基酸序列大都包含3个保守基序,分别为DNPNG、FRDP和WECPD,表明不同种属中同一基因家族的进化保守性;由聚类分析推测,VIN亚族基因功能可能与棉花纤维发育相关。 The normal development of cotton fiber requires a lot of sucrose supply,and invertase is key enzyme involved in the body of sucrose metabolism.In this study,to obtain better understanding of the quantity and distribution of sucrose converting enzymes in cotton genome,the 17-invertase family genes was used as reference sequence in Arabidopsis to determine the cotton genome database in Gossypium barbadense,Gossypium hirsutum,Gossypium raimondii and Gossypium arboreum.The results showed that the number of sucrose invertase gene family contained in Gossypiumbarbadensewas significantly higher than that in Gossypium hirsutum,while there was only one more in Gossypium raimondii than that in Gossypium arboreum.The numbers of invertase gene family members of these cultivars were 65,46,26 and 25,respectively.By analyzing the gene structure of members of the invertase gene family,αsubgroup of neutral/alkaline invertases and acidic invertase gene were mostly consisted of 6 or 7 exons andβsubgroup were consisted of 4 or 5 exons.In addition,a long UTR area was existed in the genetic structure of some Gossypium barbadense and Gossypium raimondii,but not in Gossypium hirsutum and Gossypium arboreum;The acidic invertase basically contained three conserved motifs,namely DNPNG,FRDP,and WECPD,indicating the evolutionary conservatism of the same gene family in different species.The clustering analysis inferred that the function of VIN subfamily genes might be related to cotton fiber development.These results laid a theoretical foundation for further research on cotton fiber development genes in invertase family.
