摘要
目的:采用GC和HPLC法测定并评价注射用黄芪多糖(APS-P)组成及单糖组成比。方法:以APS-P为研究对象,建立三甲基硅烷(Tri-Sil)衍生化-GC和1-苯基-3-甲基-5-吡唑啉酮(PMP)柱前衍生化-HPLC测定APS-P组成、单糖组成比的方法。通过方法学考察及对15批样品的分析,评价两种方法的适用性。结果:两种方法的专属性、精密度和稳定性较好,但PMP柱前衍生化-HPLC的操作和计算与Tri-Sil衍生化-GC相比更为简便。APS-P的组成较为固定,但单糖组成比在批与批之间呈现一定波动性。结论:PMP柱前衍生化-HPLC用于APS-P组成、单糖组成比分析更具优势。
Objective:To investigate GC and HPLC method for composition and proportion determination on injection of Astragalus polysaccharides(APS-P).Methods:Taking APS-P as the research object,two methods of composition and proportion determination on APS-P by trimethylsilane(Tri-Sil)derivatization-GC and 1-phenyl-3-methyl-5-pyrazolone(PMP)derivatization before column-HPLC were established.The applicability of two methods according to methodological investigation and the analysis of 15 batches of samples was evluated.Results:Two methods were both exclusive,precise and stable,while the operation and calculation of the PMP derivatization before column-HPLC was much easier than that of the Tri-Sil derivatization-GC.The compositions of APS-P were fixed while the monosaccharide composition proportion between different batches existed fluctuant.Conclusion:There are more advantages of PMP derivatization before column-HPLC for analyzing the composition and proportion of APS-P.
