摘要
为了研究夏枯草多酚超声辅助提取工艺及其抗氧化活性,在单因素试验基础上,采用响应面分析方法,优化超声辅助提取最佳工艺条件,并利用DPPH·清除法和·OH清除法评价夏枯草多酚的抗氧化活性。结果表明,夏枯草多酚超声辅助提取最佳工艺条件为:温度49℃,料液比(g/mL)1∶41,乙醇浓度32%。该工艺条件下,夏枯草多酚提取率为2.48%。抗氧化试验显示,夏枯草多酚与BHT清除DPPH·的IC_(50)分别为4.59μg/mL和27.36μg/mL;清除·OH的IC_(50)分别为5.52μg/mL和35.34μg/mL。表明夏枯草多酚具有强的清除自由基能力,即具有强的抗氧化活性。
In order to investigate the process of ultrasonic-assisted extraction and antioxidant activities of polyphone from Prunella vulgaris L.,the single-factor experiments and response surface method had been used to explore the optimal conditions for ultrasonic-assisted extraction. And the antioxidant activity of extraction polyphone also had been evaluated by DPPH · and · OH scavenging method. The results showed that the optimal conditions of ultrasonicassisted process was as follows :ultrasonic temperature 49℃,solid-liquid ratio(g/mL)1 ∶ 41 and ethanol concentration 32%. Under these conditions,the extraction ratio of Prunella vulgaris L. polyphone was 2.48%. The results of antioxidant activities study manifested that the half-scavenging rate(IC50)of extracted Prunella vulgaris L. polyphone and BHT to DPPH · were 4.59 μg/mL and 27.36 μg/mL respectively. And IC50 of polyphone and BHT to · OH were 5.52 μg/mL and 35.34 μg/mL,respectively. The results indicated that the extracted polyphone from Prunella vulgaris L. has strong scavenging free radicals ability(antioxidant activity).
