小麦野生近缘植物多药抗性基因PDR7的生物信息学分析 被引量:2

Bioinformatics Analysis of Multidrug-resistance Gene PDR7 in Wild Relatives of Wheat(Triticum aestivum L.)

摘要 小麦的野生近缘植物,尤其山羊草属中含有丰富的抗病虫基因,是小麦遗传育种的重要基因来源。针对禾谷镰刀菌入侵引起的小麦赤霉病,在山羊草属中尚未找到相应抗性基因。多药抗性基因(pleiotropic drug resistance, PDR)是ABC (ATP-binding cassette)转运蛋白基因三大亚家族中的一个,具有对生物和非生物胁迫的广谱抗性。本研究以D、C、U、M、S染色体基组的山羊草属植物为研究材料,对PDR7基因进行分离鉴定和生物信息学分析。结果表明,这些PDR7基因为半个PDR基因,具有PDR基因的典型结构域,是膜蛋白;同源序列对比和进化分析均表明,在山羊草属植物中,PDR7基因具有高度的保守性。以AePDR7为代表进行了启动子预测,表明AePDR7对禾谷镰刀菌等真菌、脱氧雪腐镰刀烯醇(deoxynivalenol, DON)、生物激素、盐胁迫、冷胁迫、干旱胁迫等均有响应元件,说明其可能是一个广谱抗性基因。 Wild relatives of wheat, especially Aegilops plants that are rich in insect-resistance genes, is an important source for wheat heredity and breeding. Yet till now, no corresponding resistance genes of wheat scab which caused by fusarium head blight has not been found. Pleiotropic drug resistance(PDR) gene is one of the three subfamiliesof ATP-binding cassette(ABC) transporter protein gene. It has broad spectrum resistance to biotic and abiotic stress. In this research, we used D, C, U, M and S chromosome basis set of Aegilops plants as materials to isolate and identify the PDR7 genes, and analyzed with bioinformatics method. The results showed that these PDR7 genes were half-size PDR gene, and they all had the typical structure region of PDR genes, which was membrane proteins. Homologous alignment and molecular evolution analysis showed that PDR7 was quite conserved in Aegilops plants. Use AePDR7 as the representative, promoter prediction showed that AePDR7 had response elements of fungi such as Fusarium graminearum, deoxynivalenol(DON), biotic stress, salt stress, cold stress, drought stress and so on which indicated that it might be a broad-spectrum disease resistance gene.
