摘要
ABCG转运蛋白在调控植物脂质的运输中具有重要作用。本研究从大豆品种黑农44中克隆到基因GmABCG1。大豆GmABCG1含有1 965 bp的开放阅读框,编码654个氨基酸。蛋白结构分析显示,该蛋白具有ABCG家族保守的结构域和跨膜结构域。进化树分析表明,大豆GmABCG1序列与其他豆科植物的同源基因亲缘性较近。实时荧光定量分析表明,大豆GmABCG1在果荚中表达水平较高,仅次于根。GmABCG1转基因拟南芥种子脂质含量测定表明,油脂和特异性脂肪酸含量显著增加,这暗示了GmABCG1可能参与调控大豆种子中脂质运输。
ABCG transport proteins play an critical role in regulating the transport of plant liquids. We cloned GmABCG1 gene from the soybean variety Heinong 44. The GmABCG1 gene in soybean contained 1 965 bp,encoding 654 amino acids. The analysis of protein structure indicated that this protein had the conserved domain and transmembrane domain of ABCG family. In addition, phylogenetic tree analysis showed that GmABCG1 sequence in soybean had close relationship with the homologus genes in Leguminous plants. Besides, quantitative real-time PCR showed that the expression level of GmABCG1 was relatively high in green pods, which was just slightly lower than that in roots. Moreover, determination of lipid content in GmABCG1 transgenic Arabidopsis seeds implied that the contents of oil and specific fatty acids significantly increased in GmABCG1 transgenic Arabidopsis, which suggested that GmABCG1 might be involved in the lipid transport in soybean.
