红三叶杂交F1代群体的鉴定及遗传多态性分析 被引量:1

Genetic Polymorphism Analysis and Identification on the Hybrid F_1 Population of Red Clover

摘要 为研究三红叶杂交F1代与亲本的遗传差异性和相似性,本试验利用不同地理来源且性状差异显著的抗白粉病红三叶新品系及感白粉病岷山红三叶品种作为亲本进行有性杂交,并利用AFLP分子标记对二者杂交形成的F1代单株进行鉴定和遗传多样性分析。结果表明:30个杂交F1代单株均为真杂种。11对多态性引物共扩增出197条清晰条带,165条多态性条带,其比率是83.75%。杂交F1代与亲本间遗传相似系数在0.55~0.91之间。阈值在0.718,将F1代单株和父母本分为3大类,第1类为红三叶新品系,第2类为岷山红三叶品种,第3类为30个杂交F1代单株,这些单株的性状均偏向于父本。本研究将为红三叶遗传图谱构建、基因定位和分子辅助育种提供科学依据。 In this study, one powdery mildew resistant parent-red clover new line and another susceptible variety-Minshan red clover were used to hybrid in this experiment, and the AFLP marker was used to identify the hybrid F1 generation and study their genetic polymorphism. Results showed that, 30 F1 plants used in this experiment were true hybrids, 197 bands were amplified and 165 polymorphic bands were included while using 11 pairs of AFLP prime. The polymorphic band ratio was 83.75%. The genetic similarity coefficient of the F1 plants and their parents ranged from 0.67 to 0.91. When the threshold value was 0.718, 32 materials were divided into 3 categories. One material was included respectively in the first and second groups, which were red clover new line and Minshan red clover. All F1 generations were included into the third group and their characteristics were much more similar to the male parent. This study laid the foundation for the construction of genetic map, red clover gene localization and molecular assisted red clover breeding.
