摘要
目的以人肝癌细胞HepG2为研究模型,探讨Rab17蛋白对其细胞增殖的影响。方法通过Western blot技术检测永生化肝细胞系(LO2)和肝癌细胞系(HepG2、SMMC-7721、Bel-7402)中Rab17的蛋白表达水平,再分别将靶向Rab17的小干扰RNA和Rab17过表达质粒转染于HepG2中,观察二者对HepG2细胞增殖以及其相关周期蛋白C-myc、Cyclin D1表达的影响。结果 Western blot结果表明,与永生化细胞系相比较,肝癌细胞系中Rab17表达水平显著降低。在HepG2中沉默Rab17的表达,可以增加周期蛋白C-myc和Cyclin D1的表达,促进其细胞增殖,而过表达Rab17则表现为对HepG2增殖的抑制作用。结论肝癌细胞中Rab17的蛋白表达水平明显低于永生化肝细胞,上调其表达对肝癌细胞的增殖有抑制作用。
Objective To study the effect of Rab17 protein on proliferation of human hepatocarcinoma cell HepG2.Methods The protein expression levels of Rab17 in different hepatic cell lines (L02,HepG2,SMMC-7721,Bel-7402)were detected by Westem blot.Small interfering RNA targeting Rab17 and Rab17 overexpression plasmid were transfeeted into HepG2,respectively, and their effects on HepG2 ce/1 proliferation and the expression of related periodic proteins C-myc and CyclinDi were observed.Results Western blot showed that compared with normal hepatocytes,the expression level of Rab17 in hepatocellular carcinoma cells was significantly decreased.Silencing the expression of Rab17 in HepG2 can increase the expression of periodic protein and promote the proliferation of liver cancer cells,while overexpression of Rab17 can inhibit the proliferation of HepG2.Conclusion The protein expression level of Rab17 in hepatocellular carcinoma cells was significant2y lower than that of normal hepatocytes,and its up-regulated expression had an inhibitory effect on the proliferation of hepatocellular carcinoma cells.
