青钱柳叶中16个成分的UPLC-QE-Orbitrap-MS定量分析

Quantitative analysis of 16 components in Cyclocarya paliurus leaf by UPLC-QE-Orbitrap-MS

青钱柳叶为胡桃科植物青钱柳Cyclocarya paliurus的干燥叶,现代研究表明,其化学成分复杂,具有多种生物活性。目前对青钱柳叶质量控制方面的研究主要集中于青钱柳叶中多糖及黄酮类成分含量测定,萜类和酚酸类成分含量测定研究较少。该文对青钱柳叶中3类16个活性成分进行含量测定研究,建立了青钱柳叶UPLC-QE-Orbitrap-MS定量分析方法,结果显示,16个成分在进样质量浓度范围内呈现良好的线性关系(R2> 0. 996),精密度RSD <3. 0%,重复性RSD <3. 0%,平均回收率为95. 20%~104. 4%。说明该方法简便准确,可用于青钱柳叶药材质量的综合评价。采用所建立的方法对不同产地12个批次青钱柳叶进行了含量测定,测定的16个成分在12个批次青钱柳叶中的含量相差数倍至数10倍,说明不同产地青钱柳叶内在质量差异显著。

Modern research showed that components in the dried leaf of Cyclocarya paliurus. had various biological activities. The current quality control research was focused on content determination of polysaccharides and flavonoids,while there were less research on quantitative analysis of terpenes and phenolic acids. In this paper,the contents of 16 components of 3 kinds in C. paliurus leaf were determined by UPLC-QE-Orbitrap-MS. The results were as following: good linear relationship of 16 analytes existed within the studied concentration rages(R2> 0. 996),and RSDs were of < 3. 0% in the precision test and replicate test,with the average recovery rates95. 20%-104. 4%,respectively. The results indicatod that the method is simple and accurate,which can be used for the comprehensive quality evaluation of C. paliurus leaf. The established method was applied to determine the contents of 12 batches of C. paliurus leaf from different areas,and the 16 analvtes contents in the samples could be different from several times to dozens times,which indicated that there might be significant quality difference in C. paliurus leaf from different areas.
