摘要
溶酶体组织蛋白酶(cathepsins, Cats)是一类存在于细胞内和细胞外的具有多种生物活性的蛋白酶,是哺乳动物体内蛋白水解的主要参与者。除了能影响细胞外基质稳态、自噬、细胞凋亡过程外,还对肾小球通透性、内皮功能和炎症具有调节作用。多种Cats表达活性的失调参与急慢性肾脏疾病的发生发展。其中溶酶体组织蛋白酶B(cathepsins B, CatB)、溶酶体组织蛋白酶L(cathepsins L, CatL)及溶酶体组织蛋白酶S(cathepsinsS, CatS)作为糖尿病肾病(diabetic nephropathy, DN)病理生理中的关键参与者受到了广泛的关注。目前,越来越多的证据表明CatB、L和S可能是诊断和治疗DN的新靶点。本文主要对CatB、L及S在DN的作用及相关机制进展进行综述。
Cathepsins(Cats) are a class of proteases with multiple biological activities that present in and outside the cell. It is a major player in proteolysis in mammals. In addition to affecting extracellular matrix homeostasis, autophagy and apoptotic processes, Cats also has a regulatory effect on glomerular permeability, endothelial function, and inflammation. Recent studies have shown that dysregulation of various Cats expression activities is involved in the development of acute and chronic kidney disease. Cathepsins B(CatB), Cathepsins L(CatL) and Cathepsins S(CatS) have attracted widespread attention as major players in the pathophysiology of diabetic nephropathy. At present, there is increasing evidence suggests that CatB, L and S may be new targets for the diagnosis and treatment of DN. This paper reviews the role of CatB, L and S in DN and the progress of related mechanisms.
