PLGF对大鼠坐骨神经离断模型神经远端恢复的影响

摘要 目的探索胎盘生长因子(placental growth factor,PLGF)对大鼠坐骨神经离断模型神经远端恢复的影响。方法实验规定大鼠左肢为实验组,右肢为空白对照组。将大鼠双肢坐骨神经离断后,于实验组外源添加定量PLGF工作液并包以人工神经鞘管维持营养微环境后缝合,对照组则只包饶人工神经鞘管后缝合,分别于手术后1,4,7,15,30 d取出双侧坐骨神经手术处行p75NGFR免疫组化染色及Bungner带计数,对比实验组与对照组的p75NGFR平均光密度值Density(mean)及Bungner带数量,反映雪旺细胞增殖迁移情况。结果术后1 d和30 d实验组p75NGFR的Density(mean)值明显高于对照组,1 d实验组的Bungner带计数明显高于对照组,差异具有显著性。结论外源性PLGF可促进大鼠离断坐骨神经远端p75NGFR表达,可促进雪旺细胞成行排列,间接反映出外源性PLGF可促进雪旺细胞增殖及迁移,缩短Bungner带形成时间,增加Bungner带形成数量,证实外源性PLGF对神经损伤修复具有积极作用。 Objective To explore the effect of placental growth factor (PLGF)on the distal recovery of sciatic nerve in rat model of sciatic nerve transection.Methods Defined the left leg of the rat as the PLGF group and the right one as the Control group.After both sides of the sciatic nerves of hind legs were transected at the upper thigh level,quantitative PLGF working solution was added to the PLGF group's wound with the epineural sheath conduit while the Control group was only wrapped by the epineural sheath conduit.Then sutured the muscles and skin carefully.The p75NGFR immunohistochemical staining was performed at 1,4,7,15 and 30 d after the surgery,then Bungner bands were counted and contrasted the Density(mean)between PLGF group and Control group.So that the migration of schwann cells can be reflected.Results The Density (mean)value ofp75NGFR in the 1 d and 30 d PLGF groups were significantly higher than that in the Control group.The number of Bungner band in the 1 d PLGF group was significantly higher than that in the Control group.There were significant difference. Conclusion Exogenous PLGF can promote the expression of p75NGFR in the distal end of sciatic nerve in rats,which can promote the schwann ceils stand in line.It indirectly reflected the exogenous PLGF can promote the proliferation and the migration of schwann cells.At the same time,while the formation time of the Bungner band was shorten,the number was increased.So the exogenous PLGF has a positive effect on the repair of nerve damage.
