摘要
目的:利用C2C12成肌细胞和大鼠骨骼肌体外实验体系,研究铁皮石斛提取物对运动性损伤修复作用的机理。方法:用不同浓度铁皮石斛水提物处理C2C12细胞,进行划痕实验以及提取细胞蛋白检测其EGFR的表达水平。通过手术取大鼠滑车上肌,用石斛水提物对其进行体外培养并检测其EGFR表达水平情况。结果:C2C12细胞划痕实验结果显示,石斛提取物处理2 h后对C2C12迁移增殖有一定促进作用,其作用效果与浓度相关。石斛提取物处理后的C2C12细胞EGFR表达有一定的增加趋势。体外培养肌肉组织结果表明,铁皮石斛水提物能显著提高肌肉组织中EGFR的表达水平。结论:实验结果表明铁皮石斛水提物通过正向调控C2C12细胞和骨骼肌组织的EGFR表达水平,起到促进骨骼肌细胞增殖的作用,可以对骨骼肌运动性损伤起到一定的修复作用。
Objective: To study the recovery effects of skeletal muscle cell injured and its molecular mechanism of dendrobium candidum extract on C2C12 myotubes and isolated rat skeletal muscle.Method: In this study,we adopted different dendrobium candidum extract on C2C12 cells cultured and scratch test,the expression level of EGFR(epidermal growth factor receptor) was examined.Isolated rat epitrochlearis muscle was incubated in a buffer containing dendrobium candidum extract and the EGFR activation and related events were examined.Result: The results of scratch test showed that dendrobium candidum extract(2 h) promoted the migration and proliferation of C2C12 cells,and its effect was related in a dose-dependent manner.And the EGFR expression level of C2C12 cells treated with dendrobium candidum extract was increased.In response to dendrobium candidum extract treatment,expression level of EGFR increased in rat epitrochlearis muscles.Conclusion: Dendrobium candidum extract stimulates C2C12 cells and skeletal muscle EGFR activity acutely,promoted the migration and proliferation of C2C12 cells,and been shown to have recovery effects of skeletal muscle cell injured.
