参花凝胶对豚鼠激素依赖性皮炎模型作用的实验研究 被引量:4

摘要 目的:探讨参花凝胶对豚鼠激素依赖性皮炎的作用。方法:将56只健康豚鼠(雌雄各半)随机分为空白对照组、模型组、空白基质组、参花凝胶低(以生药材计1. 5%)、中(以生药材计3. 0%)、高(以生药材计6. 0%)剂量组及阳性药物他克莫司组(0. 03%),每组8只。除空白对照组外,其余各组采用0. 05%丙酸氯倍他索酊剂建立豚鼠激素依赖性皮炎动物模型。造模成功后,各组大鼠涂抹给药,剂量为使液体均匀覆盖裸露皮肤即可,2次/d,连续15 d。观察实验前后各组豚鼠的皮损和病理改变,测定血清Ig E含量变化。结果:与空白对照组比较,模型组皮肤组织切片中乳头层和网织层排列最紊乱,炎症部位最多,并且血清中Ig E水平显著升高(P <0. 05),说明造模成功。与模型组比较,参花凝胶低、中、高剂量组皮肤外观好转,血清总Ig E水平降低,并且参花凝胶高剂量组与模型组比较差异有统计学意义(P <0. 05)。HE染色和硝酸银染色后镜下观察,与模型组相比,各个给药组皮肤组织均得到一定修复,损伤和炎症减少。结论:参花凝胶对豚鼠激素依赖性皮炎有缓解和治疗作用。 Objective: To explore the effect of Shenhua gel on hormone-dependent dermatitis in guinea pigs. Methods: Fifty-six guinea pigs( twenty-eight female and twenty-eight male) were randomized into six groups: the blank control group,the model group,low(1. 5%,calculated by raw medicinal materials),medium(3. 0%,calculated by raw medicinal materials) and high(6. 0%,calculated by raw medicinal materials) dose groups of Shenhua gel and the tacrolimus positive group(0. 03 %) with 8 ones in each. Except for the blank control group,the other groups were used to establish the animal model of hormone dependent dermatitis in guinea pigs. After the successful model establishment,guinen pigs in each group were smeared with relative drug and the dosage was enough to cover the bare skin evenly,twice a day and for 15 continuous days. The skin lesions,pathologic changes and Ig E in serum were observed before and after the treatment. Results: Compared with the blank control group,the model group presented obviously pathological changes in dorsal skin and significantly increased serum Ig E( P < 0. 05),suggesting the successful model establishment. Compared with those in the model group,the skin lesions were improved and Ig E levels decreased after the treatment with Shenhua gel,and there was significant difference between the model group and decoction group at high dose( P < 0. 05). The slices with HE staining and silver nitrate staining were observed under a microscope. Compared with those in the model group,the skin tissues in each treatment group were repaired to a certain extent,and the injury and inflammation were reduced. Conclusion: Shenhua gel can relieve hormone dependent dermatitis in guinea pigs.
