摘要 目的:制备脂质体包裹的人表皮生长因子(h EGF),并观察其对上皮细胞增殖与迁移的影响。方法:通过逆向蒸发法制备h EGF脂质体,体外培养人表皮细胞,细胞划痕后以不同浓度的h EGF脂质体处理12 h,观察细胞迁移距离;以4μg·ml^(-1)h EGF脂质体处理上皮细胞24 h后,MTT法观察细胞增殖。结果:h EGF脂质体较h EGF溶液稳定性更好,且对表皮细胞迁移和增殖有更强的促进作用。结论:h EGF脂质体更稳定高效,在表皮修复方面有更好的应用前景。 Objective:To prepare liposomes encapsulated human epidermal growth factor(h EGF)and observe the effects on the proliferation and migration of epithelial cells.Methods:h EGF liposomes were prepared by a reverse evaporation method.Human epidermal cells were cultured in vitro.After the cells were scratched,they were treated with different concentrations of h EGF liposomes for12 h to observe the cell migration distance.The epithelial cells were treated with 4μg·ml^-1h EGF liposomes for 24 h,and the cell proliferation was observed by MTT assay.Results:h EGF liposomes were more stable than h EGF solution and had a stronger effect on the migration and proliferation promotion of epidermal cells.Conclusion:Compared with h EGF solution,h EGF liposomes are more stable and efficient,which show better application prospects in epidermal repairment.
