摘要 目的:采用Box-Behnken响应面法筛选出排闼宗阳颗粒最佳提取工艺。方法:通过建立一种HPLC法能够同时测定排闼宗阳颗粒中绿原酸、咖啡酸、槲皮苷三种指标成分,并以其含量为指标,在单因素试验基础上,采用Box-Behnken响应面法考察提取时间、加水倍数、提取次数对三种指标成分的影响。结果:采用Design-Expert 8. 06软件对所得数据进行处理,并通过软件对三个模型进行整合,得到的最佳提取工艺为提取次数2次,提取时间1. 08 h,加水倍量8. 6倍。结论:该HPLC方法准确、重复性良好,Box-Behnken优化提取工艺的方法稳定可行,操作简单,可为深入研究及应用提供依据。 Objective: To screen the best extraction technology of Paita Zongyang granule by Bbox-Behnken response surface methodology. Methods: An HPLC method was established to simultaneously determine chlorogenic acid,caffeic acid and quercitrin in Paita Zongyang granule. Based on single factor experiments,Box-Behnken response surface methodology was used to investigate the effects of extraction time,water addition times and extraction times on the three index components. Results: The data were processed by design-expert 8. 06 software,and three models were integrated by the software. The optimal extraction process was twice extraction,1. 08 h extraction time and 8. 6 times of water addition. Conclusion: The HPLC method is accurate and repeatable. The extraction process optimized by Box-Behnken method is stable,feasible and simple to operate,which can provide basis for the further research and application.
